Effect of fluoride on protein and collagen biosynthesis in rabbit dental pulp in vitro

Abstract: – The time course for incorporation of 14C‐proline into various fractions of rabbit dental pulp in vitro has been measured. In the TCA‐soluble precursor pool a steady state level of activity was indicated upon incubation after 3 h, whereas incorporation into protien and 14C‐hydroxyproling, i.e. collagen formation, increased lineraly for 9 h, leaving off upon further incubation. A lag period of about 3 h was indicated for the appearance of high molecular weight 14C‐activity, including 14C‐hydroxyproline, in th… Show more

“…Indeed, F − -mediated decrease in protein synthesis and/or secretion has been well-documented [27], [28], [32], [60], [61], [62], [63], [64], [65], [66], [67], [68], [69], [70], [71], [72], [73]. Importantly, we demonstrated a decrease in enamel matrix transcripts during the maturation stage.…”

“…Fluoride toxication affects protein synthesis by primary causing destruction of polypeptide chains [34] and weakness of amino-acid bindings in proteins [35]. It has been reported that excessive amount of fluoride cause a reduction of protein synthesis [31] and also metabolisms of cholesterol and triglyceride in liver are affected in fluoride-exposed group [36].…”

“…It was concluded that F was an inhibitor of protein synthesis, thus inducing growth inhibition [26]. Other data report that collagen metabolism was affected by F in vivo [10] and in vitro [9]. The decrease of collagen synthesis has been explained by an impairment in cross-linking and maturation of collagen fibers associated with an increase in collagen degradation [10].…”

“…The influence of F on the dentinal matrix components, especially type I collagen, has been studied previously. In rabbit dental pulps incubated with graded amounts of F, release of collagen was inhibited by about 50% [9]. Sharma [10] described that long-term ingestion of F interferes with the biosynthesis, maturation, and degradation of rabbit skin collagen.…”
