ABstrAct:The effect of ethephon (80, 100 and 120 mg·l -1 ) and gibberellic acid (GA 3 ) (40, 300 and 1,000 mg·l -1 ) on the germination capacity (GC) and mean germination time (MGT) of European beech (Fagus sylvatica [L.]) dormant beechnuts or beechnuts pre-chilled for four weeks was determined. Compared to the control (dormant untreated seeds) or beechnuts treated with tap water no significant increase in mean GC was detected after the application of ethephon or GA 3 to dormant seeds. Conversely, both ethephon and GA 3 treatments reduced (ethephon significantly) GC when applied to beechnuts chilled for four weeks prior to treatment. The effect of the treatments on germination speed (MGT) and dormancy release significantly improved when beechnuts were chilled for four weeks prior to the application of ethephon or GA 3 . However, the effect of GA 3 on MGT of chilled beechnuts was not so distinct compared to dormant untreated seeds. Reduction in MGT was most obvious in seeds hydrated with 1,000 mg·l -1 GA 3 prior to germination.Their cold requirement time was reduced by three weeks compared to beechnuts hydrated in tap water.

The concentrations used had been previously optimized for these seeds (Nicolás et al, 1996, 1997; Farelli et al, 1997; Calvo et al 2003). Gibberellin 20-oxidase (GA 20-oxidase) is an enzyme that catalyses the last three steps in the synthesis of active GAs and is a potential control point in the regulation of GA biosynthesis. Reverse transcriptase-polymerase chain reaction with degenerated oligonucleotides conserved among GA 20-oxidases was used to isolate a cDNA clone for this enzyme in Fagus sylvatica L. seeds. This clone contains all the features and exhibits homology to GA 20 oxidases from several plant species. Expression of this clone, named FsGA20ox1, as a fusion protein expressed in Escherichia coli confirmed that it was able to metabolize [(14)C]GA(12) to [(14)C]GA(9) and [(14)C]GA(53) to [(14)C]GA(20). Analysis of FsGA20ox1 transcript levels showed similar low expression during stratification at 4 degrees C and in the presence of gibberellic acid or ethephon (compound that releases ethylene in solution), treatments proved to be efficient in breaking the dormancy of beech seeds. However, there was a drastic increase of FsGA20ox1 transcript levels in the presence of paclobutrazol (PCB), a well-known GAs biosynthesis inhibitor, or of 2-aminoxyacetic acid (AOA), an inhibitor of ethylene biosynthesis. Furthermore, the effect of AOA was reversed by the addition of GA(3) and that of PCB by ethephon. This indicates that the gene product is subjected to down-regulation by GA and ethylene, and further suggests a cross-talk gene regulation by these two hormones during the transition from seed dormancy to germination.

An enzymatically active recombinant protein kinase, previously isolated and characterized in Fagus sylvatica L. dormant seeds (FsPK1), was used to obtain a specific polyclonal antibody against this protein. Immunoblotting and immunohistochemical analysis of FsPK1 protein in beech seeds showed a strong immunostaining in the nucleus of the cells located in the vascular tissue of the embryonic axis corresponding to the future apical meristem of the root. This protein kinase was found to accumulate in the seeds only when embryo growth was arrested by application of ABA, while the protein amount decreased during stratification, previously proved to alleviate dormancy, and no protein was detected at all when seed germination was induced by addition of GA(3). These results indicate that FsPK1 may be involved in the control of the embryo growth mediated by ABA and GAs during the transition from dormancy to germination in Fagus sylvatica seeds.
