Caffeine is an interesting compound showing anticancer and protective effects upon topical administration, although its penetration through the skin is compromised by its hydrophilicity. Materials and methods: SLN-CAF were formulated by using a modification of the quasi-emulsion solvent diffusion technique (QESD) and characterized by PCS and DSC analyses. In vitro percutaneous absorption studies were effected using excised human skin membranes (i.e. Stratum Corneum Epidermis or SCE).Results: SLN-CAF were in a nanometric range (182.6 ± 8.4 nm) and showed an interesting payload value (75% ± 1.1). DSC studies suggest the presence of a well-defined system and the successful drug incorporation. Furthermore, SLN-CAF generated a significantly faster permeation than a control formulation over 24 h of monitoring. Discussion and conclusions: SLN-CAF were characterized by valid dimensions and a good encapsulation efficiency, although the active to incorporate showed a hydrophilic character. This result confirms the suitability of the formulation strategy employed in the present work. Furthermore, the in vitro evidence outline the key role of lipid nanoparticles in enhancing caffeine permeation through the skin.

Matricaria chamomilla L. extract has been used in cosmetic formulations. The aim of this study was to evaluate the antioxidant potential of chamomile extract as well as the clinical efficacy on skin hydration and mechanical properties of skin. The antioxidant activity of chamomile extract was evaluated by chemiluminescence (IC50 = 0.14 μg/mL). Stable formulations were supplemented with 0,5 cg/g α-bisabolol or cg/gwith 5,0 of Matricaria chamomile glycolic extract or with 0,01 cg/g of apigenin and applied on the volar forearm and face of 25 female subjects. Skin physiology was assessed before and after 2 hours (single application) and after a 2-and 4-week period of daily application. After a single application, all formulations increased the stratum corneum water content but only α-bisabolol and chamomile extract formulations reduced TEWL. The formulation with chamomile extract has shown the most pronounced result in the reduction of TEWL (27%). However, after 2-and 4-week application, only the extract formulation increased stratum corneum water content compared with the vehicle. Chamomile extract was effective in neutralizing free radicals and therefore presents an interesting potential in cosmetic formulations for this purpose. O extracto de Matricaria chamomilla L. tem sido amplamente utilizado em formulações cosméticas. Assim, o objectivo deste estudo foi avaliar o potencial antioxidante do extracto de camomila, bem como a eficácia clínica na hidratação e nas propriedades mecânicas da pele. A actividade antioxidante do extracto de camomila foi avaliada por quimioluminescência (IC50 = 0,14 ug / mL). As formulações foram suplementadas com a α -bisabolol de fração mássica 0,5 cg/g ou com extracto glicólico de Matricaria chamomilla L. de fração mássica 5,0 cg/g ou com 0,01 cg/g de apigenina e aplicados na parte interna do antebraço e na cara de 25 mulheres. A fisiologia da pele foi avaliada antes e após 2 horas (aplicação única) e 2 e 4 semanas de aplicação diária. Depois de uma única aplicação, todas as formulações aumentaram a água do estrato córneo, mas apenas as formulações com α -bisabolol e extracto de camomila reduziram a PTEA . Objective: The aim of this study was to investigate physical characteristics of nanostructured lipid carriers (NLCs) mixture of alpha tocopheryl acetate, cetyl palmitate, Tween 80 and propylene glycol using high shear homogenization technique on NLC preparation to predict the optimum ratio of alpha tocopheryl acetate-cetyl palmitate to produce good characteristics of NLC loaded coenzyme, higher % EE, good penetration, controlled release, and stable.Methods: Lipid characterizations were conducted by diffraction scanning calorimetry, X-ray diffraction, and Fourier transforms infrared spectrophotometry. Coenzyme Q10 concentration was measured by spectrophotometer at 275 nm. NLC characteristics based on their morphology was determined using transmission electron microscope, particle size, and its polydispersity index which were measured with Delsa Nano™ particle size analyzer. Percentage of coenzyme Q10 entrapped in NLC was determined by dialysis bag method. Coenzyme Q10 release profile was measured using with Franz cell for 12 hrs. The penetration depth of NLC coenzyme Q10 in abdominal skin of Wistar rat was determined with fluorescence microscopy using rhodamine B as marker. NLC physical stability based on minimum of particle size variation, pH and viscosity during 90 days storage. Results:The result showed that formula with ratio of cetyl palmitate-alpha tocopheryl acetate 70:30 (% w/w) produce good characteristics of NLC loaded coenzyme, higher % EE, good penetration, controlled release, and stable in 90 days storage. Conclusion:The coenzyme Q10 NLC system with cetyl palmitate and alpha tocopherol acetate as lipid matrixare characterized by small particle size, low crystallinity, spherical morphology of particle and high coenzyme Q10 entrapment efficiency. Crystal modification led to the formation of a more amorphous thereby increasing the drug entrapment
