Effect of an extract of the mushroom Agaricus blazei Murill on expression of adhesion molecules and production of reactive oxygen species in monocytes and granulocytes in human whole blood ex vivo

Abstract: We have reported that an extract of the edible officinal mushroom Agaricus blazei Murill (AbM) stimulates synthesis of pro-inflammatory cytokines in human monocytes and vein endothelial cells in vitro and reduces the extent of lethal septicemia in mice with bacterial peritonitis. In the present study on human monocytes and granulocytes in whole blood ex vivo, we studied the dynamic changes of cell adhesion molecules (CD11b, CD62L) and the production of reactive oxygen species (ROS) after stimulation with AbM. … Show more

“…Interestingly, stimulation with this mushroom extract induced a similar upregulation of CD69 (associated with activation) and CD86 (associated with antigen-presentation) as did stimulation with E.coli LPS. These findings are in line with the results from a previous investigation, where stimulation with AndoSan TM on MDCC was found to increase production of cytokines in a similar manner as stimulation with LPS, and, in some cases, in even higher quantities than did stimulation with LPS [34]. The β (or leukocyte)-integrins, i.e.…”

“…We evaluated ROS production of neutrophils using DCFH-DA and HE by FACS [26] and found that H 2 O 2 and production were significantly in- creased by exposure to ABM extract compared with control. These results are similar to a report that ROS production in granulocytes of human whole blood was increased by ABM [27].…”

“…Ex vivo studies [18] in whole blood demonstrated that AbM decreased the expression of adhesion molecule CD62L (L-selectin) and increased the expression of the b 2 integrin CD11b on monocytes and granulocytes. These molecules are crucial for respectively rolling and adherence to endothelial cells [19,20], and prerequisites for the ultimate transcapillary diapedesis of leukocytes to inflammatory foci in order to attack microorganisms.…”
