Effect of AgNO3 and aminoethoxyvinylglycine on in vitro shoot and root organogenesis from seedling explants of recalcitrant Brassica genotypes

Abstract: The presence of 1-10 μM aminoethoxyvinylglycine (AVG) or 5-30 μM AgNO3 markedly enhanced shoot regeneration from cotyledon and hypocotyl cultures of eight recalcitrant Brassica campestris and B. juncea genotypes tested. Expiants of B. campestris ssp. chinensis and ssp. parachinensis grown with a high AVG concentration (20 μM), regenerated poorly. All cytokinins tested were equally effective in promoting shoot formation, except that kinetin was inhibitory to shoot regeneration from hypocotyls of B. campestris s… Show more

“…[49,50] and Brassica sp. [51,52] .Supplementation of AgNO3 in culture media caused significant positive effect on shoot number with its best response being observed at 2 mgL -1 in subculture passage-III. In some plants, the regeneration potential of cultured cells and tissues has been reported to decrease with increasing cycle of subcultures [53] .…”

“…Depending upon the function of silver which appears (Beyer, 1979). The positive effect of Ag + ions in shoot regeneration suggests that ethylene produced by cultured explants inhibits shoot organogenesis of those explants (Chi et al, 1990). Plantlets resulted from the rooting previous stage were acclimatized and successfully transferred to the greenhouse (Fig.…”

“…However, a systematic study of in vitro culture of stem mustard is lack. It is necessary to establish the efficient regeneration system for mass micropropagation and genetic manipulation in this crop because genotypic difference in shoot regeneration existed in B. juncea (Chi et al, 1990;Mathews et al, 1985). In this study, we report the effect of different cytokinins and their concentrations on shoot regeneration of stem mustard and developed a reliable shoot regeneration system by using the most effective cytokinin.…”
