Effect of active immunization with IL‐17A on B cell function and infection risk in pristane‐induced lupus model

Abstract: Active immunization with IL-17A coupled to KLH was able to induce a high titer of neutralizing antibodies against IL-17A and inhibit B cell functions without increasing the risk of infection in a pristane-induced lupus mice model.

“…As a major component of humoral immunity, B lymphocytes play a crucial role in the pathogenesis of autoimmune encephalitis. 5 , 37 , 38 Autopsy pathology of patients with anti-NMDAR encephalitis showed that a large number of B cells and CD138+ cells were distributed in the perivascular, interstitial, and perivascular spaces, which provided evidence for the involvement of B cells in immune regulation. 26 B cell-targeting therapies, including RTX, are effective in anti-NMDAR encephalitis, especially in recurrent and severe patients.…”

“…13 The T helper (T h ) type 17 lymphocytes, that secrete IL-17, and IL-17 itself also have been associated in vitro and in vivo with B cell activation and antibody production, having direct B-cell helper properties. 12,[14][15][16] Several studies have confirmed the importance of IL-17 in the pathogenesis of autoimmune diseases in humans, and specifically AIHA. 11,12,17 In a murine model of AIHA, IL-17-secreting cells were significantly increased in the diseased mice, and targeted treatment that decreased IL-17 concentration decreased the incidence of AIHA.…”

“…Clinical trials showed that IFN-K was well tolerated and significantly reduced disease activity in SLE patients [163,164]. Vaccination therapy by targeting pathogenic cytokines such as IL-17 has also been studied as potential treatments for SLE [161,165,166]. B cell-targeted therapy has been regarded a promising therapeutic approach for SLE, and anti-CD20 monoclonal antibodies such as rituximab have been proven to be effective in SLE patients.…”
