The knowledge and understanding of the extent of genetic variation of mulberry germplasm is important for conservation and improvement. The objective of this study was to analyze the genetic diversity between and within two mulberry species widely grown in Kenya which include Morus alba and Morus indica. Five individuals per species were genotyped with 13 simple sequence repeat (SSR) markers. The SSR markers presented a high level of polymorphism and detected a total of 35 polymorphic bands and 74.47% polymorphic loci. The mean observed heterozygosity per primer was 0.3670 suggesting a high degree of variation. The analysis of molecular variance (AMOVA) revealed that 95% of the variation was found within the species and only 5% between the species. Principal coordinates analyses (PC O A) clearly distinguished three groups. It was evident from this study that the mulberry accessions did not cluster on the basis of their geographical origin, and neither according to the group of species they fall into. The study showed a close relationship between the two species and therefore mulberry improvement should target sampling more individuals within species rather than among species.

The chicozapote (Manilkara zapota) is a tropical fruit tree that occurs in two morphologically distinct populations in the Yucatan peninsula of Mexico. Forest populations consist of tall, straight trees, while swamp populations have a short, shrub‐like growth form. Swamp populations also have smaller leaves, fruit and seeds. We performed a random amplified polymorphic DNA (RAPD) analysis on four different populations of chicozapote to test if there was a genetic component to this variation. The populations differed in respect to habitat type (swamp vs. forest) and geographical location (east vs. west). We surveyed 80 random primers, nine of which revealed interpopulation band differences (28 band differences in total). Unweighted pair group method analysis (UPGMA) and neighbour‐joining dendrograms showed no separation of individuals between the different populations. Analysis of the RAPD data showed no significant differences between swamp and forest populations (P > 0.1). The lack of genetic differentiation suggests a failure to find a correlation between the RAPD loci and adaptive traits. The observed morphological differences between the swamp and forest populations of chicozapote may either be that gene flow has prevented a build‐up of neutral marker differences or a plastic response to differences in habitat.

In situ and ex situ assessment of morphological and fruit variation in Scandinavian sweet cherry

Sweet cherry is a tall, deciduous tree producing stone fruits. This diploid outcrossing species was domesticated in Asia but has been grown in orchards and home gardens in Scandinavia for many years. In situ and ex situ assessments of phenotypic variation in sweet cherry accessions were performed to determine the reliability of such assessments, and to determine relationships between Nordic populations. Principal component analysis (PCA) based on in situ data revealed that accessions were mostly clustered according to their country of origin. PCA based on ex situ assessment of accessions that were propagated by seed at Hornun (Denmark) did not agree with the PCA based on in situ data. These contrasting results suggest that phenotypic assessment in sweet cherry depends on the environment, genotype, and the interaction between them. Phenotypic diversity accounted for by in situ assessment may not be always true, while phenotypic differences determined by ex situ assessment may be confounded by the genotype-by-environment interaction, or could depend on the new genotypes arising from open pollination after seed propagation. Our research also suggests that ecotype differentiation could occur in wild Scandinavian sweet cherry. Fruit descriptors were among the best to distinguish between Scandinavian populations. Previously reported monogenic characteristics showed intermediate narrow-sense heritability, as suggested by the percentage of total variation accounted by the half-sib populations.
