Dual-Sensitive Graphene Oxide Loaded with Proapoptotic Peptides and Anticancer Drugs for Cancer Synergetic Therapy

Abstract: A dual-sensitive drug delivery system (DDS) based on graphene oxide (GO) which is simultaneously loaded with proapoptotic peptides and anticancer drugs was rationally designed and fabricated for cancer synergetic therapy. Specifically, a kind of cell apoptosis peptide (KLAKLAK) 2 (KLA) was anchored on the surface of GO via a disulfide bond to obtain GO-SS-KLA. Then, the aromatic anticancer drug doxorubicin (DOX) was loaded on GO through π−π conjugation and hydrogen bonding interactions. Finally, bovine serum a… Show more

“…The GO carrier was coated by bovine serum albumin (BSA) and the DOX@GO-SS-KLA/BSA DDS, which was stable in biological media, was prepared. The results showed that the reduction and pH inside the cells, respectively, caused the release of KLA and DOX, and synergy was achieved [154].…”

“…That the improvement of drug efficacy was achieved by the loading of GO that has a low toxicity has been reported [37]. Zhang et al [38] confirmed that the GO-loaded pro-apoptotic polypeptide drug delivery system effectively inhibit the proliferation of cancer cells. Similar results were reported in this study that AWRK6/GO was more efficient and effective with regard to the amelioration of the survival time and survival rate of LPS mice than AWRK6.…”

“…As the simultaneous loading of several drugs with different mechanisms of action on the same nanocarrier, obtaining a nanocarrier sensitive to double/multiple endogenous stimuli is also a way to increase therapeutic efficacy. Recently, Zhang J. et al (2019) designed and constructed a dual-sensitive cancer combination treatment system utilizing GO to load proapoptotic peptide (KLA) and anticancer drugs. The proapoptotic peptide is an amino acid sequence of lysine (K), leucine (L), and alanine (A), (KLAKLAK) 2 , that can induce mitochondrial-dependent apoptosis while remaining relatively non-toxic extracellularly.…”
