Downregulation of MACC1 inhibits invasion, migration and proliferation, attenuates cisplatin resistance and induces apoptosis in tongue squamous cell carcinoma

Abstract: The aim of the present study was to investigate the expression and function of metastasis-associated in colon cancer 1 (MACC1) in tongue squamous cell carcinoma (TSCC) and its relationship with the expression of extracellular matrix metalloproteinase inducer (EMMPRIN) (CD147). Levels of MACC1 and EMMPRIN expression were analyzed in 65 paraffin‑embedded tissue specimens of TSCC and in the adjacent non-cancerous tissues using immunohistochemistry (IHC). MACC1 expression was highly associated with lymphatic metas… Show more

“…MACC1 is known to induce proliferation, migration, and invasion in cell culture . We confirmed this observation by showing that the downregulation of MACC1 through siRNA transfection induced G1 phrase arrest, suppressed cell proliferation, migration, and invasion ability.…”

“…According to literature, MACC1 overexpression has been found in head and neck cancers, specially in tongue squamous cell carcinoma [13]. Results of our experiments showed an induced MACC1 expression of head and neck CSC; even in conmparison with non-CSC cancer cells it showed a two-fold increase.…”

“…In conformity with the result we examined if this gene could be a competitor gene for chemotherapy. According to literature, the MACC1 gene shows a clear expression in head and neck cancer cells [1]. Here we examined MACC1 expression in CSC and investigated it as a possible biomarker.…”

“…Lymph node and distant metastasis are the most reliable adverse prognostic factors in TSCC patients . Despite recent advances in its diagnostic techniques and therapeutic managements, the number of deaths associated with TSCC increased by over 10% during the past 5 years . It is, therefore, of paramount importance to understand the molecular pathways of carcinogenesis or progression to develop novel therapeutic strategies of TSCC.…”
