Downregulation of Col1a1 induces differentiation in mouse spermatogonia

Abstract: Col1a1 (one of the subunit of collagen type I) is a collagen, which belongs to a family of extracellular matrix (ECM) proteins that play an important role in cellular proliferation and differentiation. However, the role of Col1a1 in spermatogenesis, especially in the control of proliferation and differentiation of spermatogonial stem cells (SSCs), remains unknown. In this study, we explored effects of downregulation of Col1a1 on differentiation and proliferation of mouse spermatogonia. Loss-of-function study r… Show more

“…Inhibited proliferation of tumor cells is the key to apoptosis, invasion, and migration (Shimizu et al, 2013). Studies have shown (Terova et al, 2013) that the ABCE1 gene is an inhibitor of RNase L. RNase L inhibits the interferon-regulated 2-5A/RNase L pathway, which is used by viruses, influences the biological metabolism of normal cells, and induces apoptosis (Chen et al, 2012). Moreover, it can also promote cell proliferation, differentiation, and protein synthesis.…”

“…It has a long incubation period and a slow growth rate, but it often features a single metastasis at an early stage. Studies have shown that the proliferation, invasion, and migration of the tumor are closely related to gene regulation, which could provide a novel and effective means of tumor treatment (Chen et al, 2012). Studies have also shown (Pisareva et al, 2011) that the ATP-binding cassette E1 (ABCE1) protein, which is a member of the subfamily of ATP-binding cassette transporters, can inhibit cell apoptosis by interfering with intracellular ribonuclease L (RNase L) activity, and promote cell proliferation and differentiation.…”

“…Its protein product collagen-associated proteoglycan, which belongs to an extracellular matrix family, was up-regulated at the active/chronic inflammatory processes that accompany tissue damage. 30 Matrix metalloproteinases that cleave interstitial collagens also play a crucial role in regulating matrix remodeling. 31 MMP14 (MT1-MMP) is a pericellular type I collagenase that is critical for the postnatal development of the mesenchyme.…”

“…In mice, it was observed that the expression of col1a1 occurs exclusively in spermatogonia, since to the extent that these cells follow their normal course of differentiation, they lose the marking for this protein, in addition, the reduction of the expression of this protein stimulates the differentiation of the spermatogonia. In this way, the overexpression of this protein could change the differentiation of germ cells (18,19).…”
