Dose-dependent in-vivo toxicity assessment of silver nanoparticle in Wistar rats

Abstract: This study aims to suggest the limits of silver nanoparticle (AgNP) uses for medicinal purpose and was performed to explore the effect of various doses of silver nanoparticle in rats. Four different doses of AgNP (4, 10, 20, and 40 mg/kg) were injected intravenously. For safety evaluation of injected AgNP, body weight, organ coefficient, whole blood count, and biochemistry panel assay for liver function enzyme (AST, ALT, ALP, and GGTP), comet assay, ROS, and histological parameter were performed; 10-12 week ol… Show more

“…Our study aimed to analyze the toxicity of AgNPs at different doses in a murine model and to study the application of AgNPs for targeting murine fibrosarcoma. Tiwari et al 19 in 2011 studied the toxicity of AgNPs in the doses of 4 mg, 10 mg, 20 mg and 40 mg/kg b.w. in Wistar rats and concluded that doses near 4 mg/kg body weight were best suited for the therapeutic application.…”

“…in Wistar rats and concluded that doses near 4 mg/kg body weight were best suited for the therapeutic application. 19 The low dose range of 1-10 mg/kg b.w. has thus been prioritized to avoid the higher doses that would have been obviously toxic.…”

“…Tiwari et al (36) assessed increased single and double DNA breakage in rats after intravenous injection of 40 mg kg -1 bw of Ag NPs. In the second in vivo study, zebrafi sh were treated with oral Ag NPs (5 nm to 20 nm), which resulted in high levels of γ-H2AX -a marker for double DNA strand breaks.…”

“…Apart from these, cellular porous permeability was found indirectly proportional to volume of experimental colloid and time of exposure [7]. Thorny tiny silver nano particle with breadth and diameter below 1.55 to 19.57 nm range after specific experimental application found to be initiative of certain chronic kidney inflammatory diseases, [8] nano metal of this category also become barrier in proper development of viruses, fungus and certain dangerous parasites. Small size nano particles tremendously heals damaged tissue after hurts from fire injury, traumatic injury and certain diabetic and diseased wound but the negative feedback is after smooth absorption from skin it causes indirect damage to internal organs like liver and kidney followed by chronic deposition [9].…”
