This study aims to determine the diversity of culturable thermophilic bacteria isolated from eight terrestrial hot springs in Northeastern of Algeria using the conventional methods, SDS-PAGE fingerprinting of whole-cell proteins and 16S rRNA gene sequencing. In addition, their hydrolytic enzyme activities were also investigated. A total of 293 strains were isolated from the hot springs' water and sediment using different culture media. Overall, five distinct bacterial groups were characterized by whole-cell protein pattern analysis. Based on the 16S rRNA gene sequencing of 100 selected strains, the isolates were assigned to the following three major phyla: Firmicutes (93%), Deinococcus-Thermus (5%), and Actinobacteria (2%), which included 27 distinct species belonging to 12 different phylotypes, Aeribacillus, Aneurinibacillus, Anoxybacillus, Bacillus, Brevibacillus, Geobacillus, Laceyella, Meiothermus, Saccharomonospora, Thermoactinomyces, Thermobifida, and Thermus. The screening for nine extracellular enzymes showed that 65.87% of the isolates presented at least five types of enzyme activities, and 6.48% of strains combined all tested enzymes (amylase, cellulase, pectinase, esculinase, protease, gelatinase, lipase, lecithinase, and nuclease). It was found that Bacillus, Anoxybacillus, Aeribacillus, and Aneurinibacillus were the genera showing the highest activities. Likewise, the study showed an abundant and diverse thermophilic community with novel taxa presenting a promising source of thermozymes with important biotechnological applications. This study showed that a combined identification method using SDS-PAGE profiles of whole-cell proteins and subsequent 16S rRNA gene sequence analysis could successfully differentiate thermophilic bacteria from Algerian hot springs.

Bacterial biodiversity in mineral medicinal waters and the antibiotic resistance profiles, is a developing topic in Ecuador. The objective of the work was to know the bacterial microbiota and its resistance profiles to antibiotics of the mineral medicinal waters of the Santagua Chachimbiro spa, located in the Province of Imbabura-Ecuador. 16 water samples were collected. The isolation of the bacterial species was carried out by the membrane filtration technique, using different types of culture media. The identification was carried out according to the schemes proposed by MacFaddin (2003), complemented with the tests of the Microgen galleries. The antibiotic resistance profile was determined by the Kirby and Bauer (1966) plate diffusion method. 85 strains were isolate of which 61% were Gram negative and 39% Gram positive. The main species identified were Aeromonas caviae, Aeromonas eucrenophila, Aeromonas hydrophila, Aeromonas media, Aeromonas salmonicida, Aeromonas schubertii, Bacillus subtilis, Bacillus mycoides, Bacillus spp, Burkholderia cepacia, Citrobacter freundii, Comamonas spp, Pseudomonas aeruginosa, Pseudomonas fluorescenscens, Pseudomonas putida, Pseudomonas stutzeri, Ralstonia pickettii, Staphylococcus aureus, Staphylococcus epidermidis, Staphylococcus lugdenensis, Staphylococcus saprophyticus, Staphylococcus spp, Staphylococcus warneri and Staphylococcus xylosus. Regarding antimicrobial resistance, most of the Gram negative strains were resistant to penicillin and cephalosporins. Gram positive to oxacillin and penicillin 66,67% were multiresistant to more than three antibiotics. The “Termas de Santagua de Chachimbiro” spa presents a diversity of bacterial species and the presence of environmental resistomes. Contamination by lead (Pb) has become a serious health and environmental problem, that has to be urgently prevented. Bioremediation is one of the environmentally friendly methods for eliminating Pb from the contaminated environment. This study was aimed to investigate the potency of lead-tolerant thermophilic bacteria isolated from solfatara Mount Merapi, Indonesia. A total of 340 isolates of thermophilic bacteria were screened for lead tolerance at 55 °C. Five bacterial isolates were found to show tolerance to 100 mg/L Pb (II), and then were further evaluated and identified as Aeribacillus pallidus strains MRP112, MRP148, MRP272, MRP278, and MRP280 based on 16S rRNA gene sequences analysis. Among the five isolates, A. pallidus MRP 280 showed highest activity in removing Pb at pH 6, 55°C for 24 h. The analysis of scanning and transmission electron microscopy, biofilm formation, and siderophore production, demonstrated that lead tolerance of A. pallidus MRP 280 strain was also accompanied by morphological changes, bioaccumulation, biosorption, biofilm, and siderophore assembly. In conclusion A. pallidus MRP 280 was demonstrated as one of the most potential bacterial strains, can be recommended as an agent for high-temperature lead bioremediation.
