Distribution of Doxorubicin in the Bladder Wall and Regional Lymph Nodes After Bladder Submucosal Injection of Liposomal Doxorubicin in the Dog

Abstract: The present results demonstrate that Lip-Dox injected into the bladder submucosally distributes well, both in the whole bladder wall and in regional lymph nodes and remains at a high concentration in these tissues for at least one week after injection.

“…Submucosal injection of doxorubicin-loaded liposome in bladder was shown to result in better distribution of drug and prolonged retention through the bladder wall and regional lymphoid nodes compared to free drug. The result suggests that the therapeutic use of nanocarrier not only in superficial bladder cancer but also in invasive bladder cancer and regional lymphoid node metastasis33.…”

“…Another alternative administration method was investigated in dogs that were injected with lipo‐DOX in the submucosal bladder layer with cystoscopic guidance . DOX levels were subsequently measured in the bladder wall and regional lymph nodes and were found to be significantly higher at both these sites compared with dogs that were treated with free DOX intravenously.…”

“…The results from the preclinical lipo‐DOX submucosal bladder injection study seemed promising for increasing local control as well as targeting regional lymph node metastasis . It would be interesting to see the potential therapeutic benefit as well as toxicity in a spontaneous canine cancer model, and whether this would pose a benefit compared with current standards.…”

“…[20] Submucosal injection of liposomes loaded with doxorubicin showed better epithelial drug retention as well as regional nodal uptake in bladder tumors. [21] This may provide a therapeutic alternative in invasive bladder tumors. Coumarin-loaded polylactide-co-glycolide nanoparticles were modified with the addition of poly guanidinium oxanorbornene.…”
