Ethnomedicine reflects the fusion of tradition, natural resources, and local knowledge of the community in maintaining health. However, this knowledge is facing the threat of disappearance due to poor documentation and socio-cultural alteration. This study was conducted to find out information about species of plants used and related practices by traditional healers to overcome health problems. The study was conducted in Rongkong, North Luwu District, South Sulawesi Province. A semi-structured questionnaire was developed to obtain informants demographic data, species of medicinal plants, traditional medicine ingredients, parts used, methods of remedies preparation and use, as well as various ailments that can be treated by medicinal plants. The data were analyzed descriptively and quantitatively, by calculating the family and species use-value. The study revealed medicinal plants used and practices by Rongkong traditional healers to treat 31 ailments. They used a total of 62 species of medicinal plants distributed among 33 botanical families and 55 genera. Allium cepa has the highest UV among all medicinal plants in Rongkong. Both traditional knowledge and medicinal plant diversity possessed by Rongkong community are valuable local assets that must be maintained and developed for greater benefits.

Local people's trust in traditional medicine methods is still high, especially in rural areas. Most of the villagers in Tulakan Sub-district, Pacitan District, East Java, Indonesia, grow their medicinal plants in their yards. However, local people's knowledge of various medicinal plant types is only conveyed orally from parents to children and community practice habits. Information about the using medicinal plants may begin to degrade from time to time. This study aimed to investigate the knowledge of local communities regarding the use of medicinal plants and the plant diversity, especially in the village of karst areas. The research was conducted in Bungur Village and Tulakan Village, Tulakan Sub-district, Pacitan District, East Java, Indonesia. Data collection was carried out using the snowball technique through open interviews and field surveys. A total of 40 respondents with 7 people of young age range 15-40 years and 33 people of old age range 41-75 years. Respondents with the most educational backgrounds are high school graduates. A total of 51 species were found, from 46 genera and 29 families, with 35 diseases being treated. Fever, cough, and external wounds are the most commonly treated diseases. The most widely used medicinal plant is Zingiber officinale Roscoe to warm the body, prevent fever, and increase immunity. During the COVID-19 pandemic, the villagers orally consumed ginger for post-COVID-19 positive recovery and maintained health. The most used plant parts are leaves and rhizomes by decoction and consumption orally. Based on the research results, it is known that the knowledge of the local communities and the use of medicinal plants is still quite good among the older and younger people with various types of plants.

Kunyit putih merupakan tanaman obat yang bermanfaat dan perlu dibudidayakan lebih luas namun terkendala oleh penyediaan bibit bermutu yang terbatas. Perbanyakan tanaman dengan cara konvensional yang dilakukan turun temurun dan terinfeksi patogen menyebabkan kualitas rimpang menurun. Perbanyakan secara in vitro dapat mengatasi kelangkaan bibit kunyit putih yang bermutu tinggi dalam jumlah besar dan bebas patogen. Ketersediaan bibit unggul yang dihasilkan secara in vitro bergantung pada jenis dan konsentrasi sitokinin yang ditambahkan pada media kultur. Tujuan penelitian adalah untuk menetapkan jenis dan konsentrasi sitokinin yang paling baik pengaruhnya terhadap pertumbuhan tunas kunyit putih (Kaempferia rotunda L.) secara in vitro. Percobaan dilakukan di Laboratorium Kultur Jaringan, Fakultas Pertanian, Universitas Padjadjaran yang berlangsung pada bulan Agustus 2019 – April 2020. Bahan tanam berasal dari tunas rimpang yang berukuran ±0,5-1,0 cm. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) 10 perlakuan dengan 3 kali ulangan. Media yang digunakan adalah Murashige and Skoog (MS) dengan perlakuan jenis sitokinin Benzil Amino Purine (BAP), 2-Isopenteniladenina (2-iP) konsentrasi 1.5; 3.0; dan 4.5 ppm dan Thidiazuron (TDZ) konsentrasi 0.15; 0.30; 0.45 ppm. Hasil percobaan menunjukkan terdapat pengaruh yang berbeda dalam memacu dan merangsang pemunculan jumlah tunas, tinggi tanaman, dan panjang akar pada tunas kunyit putih secara in vitro. Pemberian 0.45 ppm Thidiazuron pada media MS memberikan pengaruh terbaik dalam menghasilkan jumlah tunas. Kata Kunci: Tunas Rimpang, Perbanyakan, Kaempferia rotunda, Sitokinin
