Differentiation of thymocytes during chicken ontogeny: occurrence of a specific DNA ligase in relationship to cell size and surface antigens

Abstract: The activities of two forms (8.2 and 6.2 S) of DNA ligase have been studied in chicken thymocytes at different stages of development (16 and 18 days of incubation, hatching and one month after birth). Thymocytes have been sorted out on the basis of size and T antigenicity (immunofluorescence) with the activated cell sorter. Three kinds of cells can be distinguished: (a) large, antigenically negative cells of 16 days of incubation, expressing only the "heavy" (8.2 S) form of enzyme. The activity of this form de… Show more

“…It was also observed that 6 S DNA ligase is expressed only in mature thymocytes characterized by the expression of a T cell differentiation marker [18]. As demonstrated by Le Douarin and co-workers [12,131 the bird thymus is colonized during the embryonic and perinatal life by successive waves of LPC.…”

“…For sorting by fluorescence-activated cell sorter (FACS), cells were labeled by indirect immunofluorescence assay with anti-T or anti-B xenoantisera as previously described [15], or with anti-B mAb according to the procedure of Forni [18]. Cell sorting was performed on FACS I1 (Becton Dickinson, Sunnyvale, CA) according to cell size or fluorescence as already described in a previous report [15].…”
