Differential gene expression in colon carcinoma cells and tissues detected with a cDNA array

Abstract: Expression of selected genes coding for proteins with defined cellular functions was analysed in human cell lines derived from normal colonic mucosa, non‐mucinous colonic carcinomas and mucinous colonic carcinomas. Altered expression of 10 genes in colon carcinoma cells was found by using a cDNA array; 6 of these alterations (60%) were confirmed by Northern blotting or semi‐quantitative reverse transcription‐polymerase chain reaction (RT‐PCR). Among these 6 genes, 3 transcription factors as well as the topoiso… Show more

“…This ratio is in agreement with other reports (Zhang et al, 1997;Stremmel et al, 2002). Comparison with lists of discriminator genes previously identified in CRC using DNA microarrays (Alon et al, 1999;Backert et al, 1999;Kitahara et al, 2001;Notterman et al, 2001;Agrawal et al, 2002;Birkenkamp-Demtroder et al, 2002;Lin et al, 2002;Zou et al, 2002;Frederiksen et al, 2003;Tureci et al, 2003;Williams et al, 2003) revealed many common genes, further emphasizing the validity of our data. For example, CA4, CHGA, CNN1, MYH11, FCGBP, KCNMB1 and SST were downregulated, whereas CA3, CCT4, EIF3S6 or EEF1A1, IFITM1, CSE1L, NME1 or RAN were upregulated in cancer samples.…”

“…Gene expression profiling-based studies of CRC have so far compared normal and tumour tissue samples or different stages of disease (Alon et al, 1999;Backert et al, 1999;Hegde et al, 2001;Kitahara et al, 2001;Notterman et al, 2001;Agrawal et al, 2002;Birkenkamp-Demtroder et al, 2002;Lin et al, 2002;Zou et al, 2002;Frederiksen et al, 2003;Tureci et al, 2003;Williams et al, 2003), but none has directly addressed the issue of prognosis or MSI phenotype. Here we have applied DNA microarray technology to analyse the expression of B8000 genes in 50 cancer and noncancerous colon tissue samples.…”

While b-actin was expressed at comparable levels in all groups of tumors, GnT-IV and Nup50 were dramatically upregulated in the metastasizing primary tumors and in the correspondent metastases.…”

“…Mainly advanced cancer has been analysed in gene expression profiling-based studies on colorectal cancer (Alon et al, 1999;Backert et al, 1999;Hegde et al, 2001;Kitahara et al, 2001;Notterman et al, 2001;Takemasa et al, 2001;Agrawal et al, 2002;Birkenkamp-Demtroder et al, 2002;Lin et al, 2002;Zou et al, 2002;Frederiksen et al, 2003;Muro et al, 2003;Tureci et al, 2003;Williams et al, 2003;Bertucci et al, 2004). Only a small number of adenoma and early invasive cancer tissues have been analysed in previous studies, and the issue of flat-and protruded-type adenoma tissues has not been directly addressed (Notterman et al, 2001;Agrawal et al, 2002;Lin et al, 2002;Williams et al, 2003).…”
