Gene expression profiling of colorectal adenomas and early invasive carcinomas by cDNA array analysis

Abstract: It is generally accepted that most colorectal carcinomas arise in pre-existing adenomas. Morphologically, colorectal adenomas can be divided into two groups, protruded type and flat type. The aim of this study was to clarify relevant alterations of gene expression associated with the early stage of colorectal carcinogenesis. Using cDNA array, we analysed the expression profiles of 550 cancerrelated genes in 36 colorectal adenomas (18 flat-type and 18 protruded-type adenomas) and 14 early invasive carcinomas. A… Show more

“…Furthermore, coinciding data was shown in other reports in which fresh samples were used without amplification. For instance, HLA-DQA1 was also reported to be down-regulated in colon adenomas (10). Similarly, genes such as PDCD4, SEPP1, TSNAX and IGLJ3 were also downregulated in our previous expression profiling study of stomach cancer without amplification (11).…”

“…It was of interest that down-regulation of HLA-DQA1 has also been described by Nosho et al using fresh tissue samples of colon polyps without signal amplification (10). It has been implicated in Table III.…”

“…Adenomatous colorectal polyps are one of the most common human tumors and the most frequent precancerous lesions in the colorectum, but their transcriptome has been only partially analyzed, and the data are generally based on a limited number of cases (17)(18)(19)(20). We attempted to fill this gap by doing a comprehensive whole-genome microarray analysis of a large, highly homogenous set of adenomas that was collected prospectively.…”

“…Evidence suggests that BMPs are important in tumorigenesis (8)(9)(10). BMPs are members of the transforming growth factor beta (TGFβ) family; thus, BMPs utilize a similar signaling cascade to that of TGFβ.…”
