Dietary regulation of adiponectin by direct and indirect lipid activators of nuclear hormone receptors

Abstract: Adiponectin is an adipokine mainly secreted by adipocytes that presents antidiabetic, anti-inflammatory, and antiatherogenic functions. Therefore, modulation of adiponectin expression represents a promising target for prevention or treatment of several diseases including insulin resistance and type II diabetes. Pharmacological agents such as the nuclear hormone receptor synthetic agonists like peroxisome proliferator activated receptor γ agonists are of particular interest in therapeutic strategies due to thei… Show more

“…The beneficial effects of magnesium intake on blood lipid parameters may be explained by the regulatory roles on the activity of lipoprotein lipase, fatty acid desaturase and lecithin‐cholesterol acyl transferase which are involved in the maintaining lipoprotein balance and fatty acid oxidation . Moreover, vitamin E induces adiponectin expression which involved in the regulation of glucose and lipid metabolism . In addition, vitamin E mediates carbohydrate metabolism through the increasing of intracellular magnesium and glutathione levels …”

“…9CDHRA can activate RXR and via this route also initiate nonpermissive heterodimers, such as RXR-PPAR, RXR-liver X receptors (LXRs), RXR-farnesoid receptors (FXR) and RXR-nuclear hormone receptor 4A (NR4A) protein, involved in the expression of a wide array of genes involved in inflammation and lipid metabolism, as reviewed in Desvergne (161) . A large overlap was found between beneficial anti-inflammatory effects of carotenoids in general and lipid metabolism for many RXR-heterodimer-mediated signalling pathway targets (131,(161)(162)(163)(164)(165)(166) .…”

“…This adipokine has insulin-sensitisation properties of the target cells (liver, skeletal muscle and adipose tissue) and can enhance glucose uptake in muscle tissue (11)(12)(13) . Adiponectin also protects β-cells from immune attack and reduces insulin resistance through immunoregulatory and anti-inflammatory effects (11)(12)(13)(14) . In pregnant women, concentrations of adiponectin have been inversely associated with adverse maternal outcomes, such as GDM (15,16) , overweight and obesity (17) .…”

“…Many factors can influence the expression of this adipokine. Vitamin D concentration is one example (13,20) , as it down-regulates pro-inflammatory cytokines and up-regulates anti-inflammatory cytokines. Vitamin D appears to be positively associated with adiponectin gene expression and suppresses the renin-angiotensin-aldosterone system (RAAS) components secreted by adipocytes, thus contributing to increased adiponectin secretion (20)(21)(22) .…”
