Metabolomics focused on reproduction have been the subject of special interest in the past decade. Metabolite profiling provides detailed information on the status of follicular fluid and follicular cells which accompany the growing oocyte. Although nutrients present in the diet reach oocytes via the follicular fluid, it is not evident whether oocyte/embryo quality can be predicted based on the follicular fluid composition. Since this phenomenon has not yet been investigated in the pig, the aim of the present study was to investigate associations between diet composition and (1) selected markers related to developmental potential of oocytes (brilliant cresyl blue -BCB test, relative transcript abundance of EEF1A1 and ATP5A1 marker genes) and (2) fatty acid profile in the follicular fluid. Gilts were fed control and experimental diets which differed in composition and fatty acid profiles. The experimental diet used in this study comprised mainly locally grown grains (barley and rye) traditionally used for fattening pigs in Poland. Cumulus-oocyte complexes were aspirated from individual pairs of ovaries after animal slaughter, evaluated morphologically and subjected to the BCB test. Relative transcript abundance for the two marker genes was monitored by real-time PCR in oocytes of both categories (BCB + and BCB -). Fatty acid profile in follicular fluid was analyzed by gas chromatography. We show that the experimental diet rich in n-3 fatty acid significantly influences fatty acid composition of the follicular fluid. The fatty acid profile of the follicular fluid of gilts fed the experimental diet differed from that of the control females. The content of saturated fatty acids was higher in the experimental group, whereas unsaturated and polyunsaturated fatty acids were more abundant in the control group (P < 0.05). With regard to individual fatty acids, only C16:0 (palmitic acid), C18:2n-6 (linoleic acid) and C22:6 (docosahexaenoic acid) differed significantly. The abundance of ATP5A1 mRNA was influenced neither by diet composition nor by oocyte category (BCB + /BCB -) whereas that of the EEF1A1 was affected by both factors. Since higher mRNA level of the EEF1A1 gene was noted in BCB + oocytes this may be considered as a marker of oocyte quality in the pig. Maternal nutrition is one of the main factors affecting developmental competence of mammalian oocytes and embryos. Diet composition may alter the maternal environment of the oocyte and embryo growth through metabolic and endocrine modifications. Zak et al. (1997) showed that feed intake influenced the meiotic competence of porcine oocytes. They claimed that maternal under-nutrition may reduce the follicle's ability to support maturation of oocytes and thus their quality. Resumo -O objetivo deste trabalho foi avaliar o efeito da suplementação alimentar com gordura protegida ruminal sobre as estruturas ovarianas e sobre a concentração sérica de progesterona, em novilhas Nelore mantidas em pasto. Quarenta novilhas foram divididas em dois grupos: um suplementado com gordura protegida Megalac-E (G); e outro sem suplementação de gordura (C). O grupos foram avaliados em delineamento "crossover". Utilizaram-se dietas isoenergéticas e isoproteicas. Após 15 dias de suplementação, os animais foram submetidos a um protocolo hormonal, para avaliação da influência da suplementação com gordura no metabolismo da progesterona. Para isto, em um dia aleatório do ciclo (D0), inseriu-se um implante intravaginal de liberação de progesterona (CIDR), e aplicou-se prostaglandina F2α (PGF2α, i.m.). No D7, o implante foi retirado, e outra aplicação de PGF2α foi realizada. No D18, foi feita uma nova aplicação de PGF2α e, então, foram observados diariamente os exames ultrassonográficos ovarianos e a ocorrência de estro. Para o ensaio com progesterona, colheu-se sangue 4 dias após a inserção do implante e, novamente, 7 e 14 dias após a ovulação. A concentração de progesterona sérica no D4 foi maior no grupo G. Não houve diferença nas concentrações séricas de progesterona 7 e 14 dias após a ovulação, nem no diâmetro do folículo ovulatório, nem no volume luteal. A suplementação com Megalac-E altera o metabolismo de progesterona, mas não altera a função ovariana em novilhas zebuínas em pasto.Termos para indexação: Bos indicus, ácidos graxos, corpo lúteo, folículo, progesterona. Forty heifers were divided into two groups: one supplemented with rumen-protected fat Megalac-E (F); and another without fat supplement (C). The groups were evaluated in a crossover design. The diets were isocaloric and isoproteic. After 15 days of supplementation, the animals were submitted to a hormonal protocol to evaluate the influence of fat supplementation on progesterone metabolism. For this, on a random day of the cycle (D0), the heifers received an intravaginal progesterone-releasing device (CIDR) and an injection of prostaglandin F2α (PGF2α, im). On D7, CIDR was removed and another injection of PGF2α was performed. On D18, a new treatment with PGF2α was done and, then, daily observations of estrus and ovarian ultrasound exams were carried out. The evaluation of the effect of supplements of conjugated linoleic acid (CLA) given to pregnant and lactating sows on the phenotypic manifestation of their reproductive performance and on the birth and weaning weight of piglets was performed under conditions of a commercial herd of pigs. The experiment involved altogether 40 hybrid sows divided into two equal groups, i.e. Controls with 20 sows and Experimental Group with also 20 sows. Experimental Group received within the time interval from the 108th day of pregnancy to the 28th day after the parturition a supplement of 2 % of CLA. In individual litters the following performance parameters were recorded: number of all newborn piglets, number of piglets born alive, number of stillbirths, number of weaned piglets and individual weights of newborn piglets and weaned piglets. Obtained results concerning numbers of all newborn piglets in Controls and Experimental Group (i.e. 12.20 ± 2.09 and 11.85 ±2.58, respectively) indicate that supplementing of the feeding ration with CLA in the last stage of pregnancy does not influence the size of litter. In Controls, numbers of piglets born alive were higher by 0.4 animals per litter; this difference, however, was statistically insignificant. Also the weights of newborn and weaned piglets were not influenced by the supplement of CLA. Statistically significant differences (P ≤ 0.01) were found out between Controls and Experimental Group in numbers of weaned piglets, viz. 9.75 ± 1.68 vs. 8.35 ± 1.39, respectively, and between corresponding losses until weaning, viz. 0.95± 1.39 vs. 2.60 ± 1.93 in Controls and Experimental Group, respectively (P ≤ 0.01).
