Our century is characterized by the introduction of innovative technologies that are used in all spheres of life, including in pedagogy, which helps to familiarize students with modern medical and pharmaceutical technologies. These drugs are obtained by using biological objects by means of special technological processes of cell engineering, hybrid technologies, engineering enzymology, engineering immunology, and others. Biotechnological preparations include certain antibiotics, hormonal and vitamin preparations, blood preparations, cytokines, immunomodulators, enzymes, lipids, polysaccharides, monoclonal antibodies, and others. Biotechnological preparations differ from chemical synthesis drugs not only in their production technology, but also in their significant molecular weight, complexity of the spatial structure of the protein, a certain ratio of isoforms, instability of molecules, and other properties. At the same time, they have a significant cost due to the complexity of the technological process: it is impossible to create analogues of biotechnological drugs by chemical synthesis. If desired to reproduce biotechnological products, biosimilars are obtained which must be subjected to complete preclinical (in vitro and in vivo) studies, taking into account the definition of pharmacokinetics and clinical trials. In Ukraine the exchange of biosimilars is regulated by the pharmacovigilance system. To improve the quality of teaching the pharmacology to students of medical and pharmaceutical faculties, it is necessary to modernize educational technologies. For this purpose, biotechnological preparations are included in the pharmacology program for students of medical and pharmaceutical faculties.

To develop a method for isolation and purification of recombinant human interleukin-7 from bacterial inclusion bodies. Methods. Protein synthesis via fermentation, densitometry, refolding method of gel filtration, two-stage chromatographic purification of the protein.Results. The inclusion bodies obtained via fermentation in E.coli cells contained recombinant human interleukin-7, which was solubilized at 22ºС for 1 hour in the following buffer solution: 7M of guanidine hydrochloride, 100mM of Tris-НСl, 0.1 % Tween-20 and 50mM of dithiothreitol. Renaturation was conducted by replacing the buffer on column XK26 / 40 (GE Healthcare) with Sephadex G-25 fine equilibrated Clark-Labs solution containing L-arginine hydrochloride and tween-20. Subsequent purification of rIL-7 on the gel filtration column was performed in two stages: at the first stage, purification of rIL-7 was performed on a column packed with 20 ml of Q sepharose, at the second stage, the rIL-7 fraction purified on Q sepharose was purified on SP sepharose. Elution of rIL-7 was performed using double-stage gradient NaCL (0.2 and 0.7M) in a buffer 0.05 M of KH 2 PO 4 /NaOH with pH 6.0; 0.1 М of L-arginine; 0.1 % Tween-80. Conclusions. The amount of the target protein after all the proposed stages of purification was 10 % with the purity ratio about 95 %, which is sufficient for further development of the various pharmaceutical forms.

Вступ. Гіпергомоцистеїнемія є небезпечним метаболічним порушенням, що призводить до виникнення низки захворювань.Проблематика. Нагальним завданням є розроблення препаратів, які здатні знижувати рівень гомоцистеїну, не спричиняючи побічних ефектів.Мета. Розробити дієтичну добавку, що при мінімальному вмісті компонентів, які здатні викликати побічні реакції, знижує рівень гомоцистеїну; а також дослідити, чи впливає розроблена добавка на когнітивні здібності тварин, та впровадити її у виробництво.Матеріали й методи. До складу розробленої дієтичної добавки «Альфакогнітин» включено вітаміни В6, В9, В12, С і холін. Моделювання експериментальної гіпергомоцистенемії у щурів проводили шляхом утримання тварин наL-метіоніновій дієті. Вміст гомоцистеїну у крові визначали за допомогою іонообмінної рідинно-колонної хроматографії з використанням автоматичного аналізатору амінокислот. Поведінкові реакції та когнітивні здібності щурів досліджували за допомогою поведінкових тестів «Відкрите поле», «Електрична стимуляція кінцівки» і «Соціальнавзаємодія». Роботи щодо впровадження у виробництво виконано за участі компанії ТОВ «Нутрімед» (Київ).Результати. Показано, що у тварин із гіпергомоцистеїнемією «Альфакогнітин» знижував рівень гомоцистеїну, підвищував когнітивні здібності, ефективність соціальної взаємодії та комунікабельність, а також нормалізував функціональні порушення пам’яті та здатності до навчання. Затверджено технічні умови виробництва дієтичної добавки,відпрацьовано пілотну технологію отримання її капсульованої форми та виготовлено дослідну партію.Висновки. «Альфакогнітин» може знижувати рівень гомоцистеїну, що дозволяє використовувати його з метою нормалізації функціонального стану серцево-судинної та нервової систем за гіпергомоцистеїнемії, а також для покращення когнітивних функцій, зокрема після захворювання на COVID-19.
