Medicated chewing gums (MCGs) represent a beneficial platform for realizing drugs intended for dental prophylaxis and treatment. The present study aimed to investigate the impact of compression force on the mechanical, textural, release, and chewing perception characteristics of compressible MCGs with the combination of lysozyme hydrochloride (LH) and ascorbic acid (AsA). Four batches of MCGs were obtained on a laboratory single-punch tablet machine applying different forces, i.e., 5, 7, 10, and 15 kN, and evaluated by their geometrical parameters, mechanical resistance, surface and internal structure characteristics, texture profile, release behavior, and perception attributes during mastication. It was found that increasing compression force slightly affected resistance to crushing and friability of MCGs, but resulted in surface smoothing and formation of a thicker layer with highly compacted particle arrangement. According to the texture analysis, increasing compression force led to harder and more adhesive gums, indicating possible difficulties in chewing and, therefore, impairment of their consumer properties. Lower compression forces were also found to be preferable in terms of better drug release from the obtained chewing gums. The volunteers’ assessment showed that an increase of compression force led to significantly raising the initial hardness and crumbliness as well as to decreasing the rate of the integral gum mass formation during mastication, which may negatively affect perceptive sensations when using MCGs. Based on the results obtained, the optimal compressing force was selected to be 7 kN, which allows obtaining MCGs with good organoleptic, mechanical, textural, and release properties.

Overcoming the bitter taste is a main problem in developing a pleasant product, especially for paediatric patients. According to our best knowledge, literature data does not provide testing suitability of sweeteners in masking the taste of levocetirizine dihydrochloride in gels. The present work focuses on the effect of three sweeteners (neotame, saccharin sodium and cyclohexyl sulfamate sodium) on masking the bitter taste of levocetirizine dihydrochloride in a mucoadhesive gel. The study shows not only the graphs of the formulation, active pharmaceutical ingredient (API) and placebo, but also the process of the bitter substance release from the gel as seen using the electronic tongue. In this work a potentiometric electronic tongue based on 16 ion-selective electrodes with solid contact based on silver chloride electrode was used. The principal component analysis (PCA) method was used to analyse the results statistically. Release tests were also performed. The collected samples were analysed using the reversed-phase HPLC to determine the levocetirizine dihydrochloride content in each of them. The formulations presented in the plot form compact clusters which proves the repeatability of the e-tongue responses. The electronic tongue release study suggests that all the sweeteners in the appropriate concentrations presented in this study could mask the bitter taste of levocetirizine dihydrochloride. RezumatMascarea gustului amar este o problemă principală în dezvoltarea unui produs plăcut, în special pentru pacienții pediatrici. Lucrarea de față se concentrează asupra efectului a trei îndulcitori (neotam, zaharină sodică și ciclohexil-sulfamat de sodiu) asupra mascării gustului amar al diclorhidratului de levocetirizină într-un gel mucoadeziv. Studiul prezintă rezultatele formulării ingredientului farmaceutic activ (API) și placebo, precum și procesul de eliberare a substanței amare din gel, folosind metoda limbii electronice potențiometrice. Această metodă folosește 16 electrozi ion-selectivi cu contact solid pe bază de electrod de clorură de argint. Evaluarea componentelor principale a fost utilizată pentru a analiza statistic rezultatele. Alături de testele de cedare, probele colectate au fost analizate folosind HPLC în fază inversă pentru a determina conținutul de diclorhidrat de levocetirizină în fiecare dintre ele. Formulările prezentate formează clustere compacte care demonstrează repetabilitatea răspunsurilor e-limbii. Studiul de eliberare electronică a limbii sugerează că toți îndulcitorii în concentrațiile adecvate prezentate în acest studiu ar putea masca gustul amar al diclorhidratului de levocetirizină.

The main objective of this study was to prepare and characterize the MCG of Ondan HCl which helps to improve the effectivity of the drug and to investigate the applicability of the SeDeM system for the formulation of MCG. The Ondan HCl was formulated into directly compressed MCG utilizing the SeDeM Diagram Expert System. The MCG was optimized using 32 factorial design. Performance evaluation was carried out by evaluating different parameters. Statistical analysis showed that micromeritic properties were improved after inclusion of appropriate excipient in optimum amount. In-vitro drug release study showed 90.01% drug release whereas ex vivo buccal permeation study exhibited significant drug permeation within 15 min indicating its potential for increasing bioavailability. It was concluded that various characteristics of powder material at pre formulation level can be successfully predicted by SeDeM diagram expert system. The developed medicated chewing gum is more effective than mouth dissolving and conventional tablet formulation.
