Development of a Lateral Flow Immunoassay (LFA) Strip for the Rapid Detection of 1‐Aminohydantoin in Meat Samples

Abstract: Nitrofurantoin is one of antibiotics of the nitrofuran family, which has been used not only to prevent and treat diseases, but also to promote growth in animals. However, concerning the carcinogenicity of the metabolite of nitrofurantoin (AHD), a new fast and convenient method for monitoring AHD should be established. We describe the development of a new test assay for rapid screening of meat samples.

“…Based on the results from 20 different blank animal tissue samples (chicken, pork, carp and shrimp), the LODs and LOQs of ICTS were 0.15 and 0.31 μg/kg, respectively. The LOD and LOQ values achieved in this study were lower than those in published papers (Li et al, 2015;Tang, Xu, Liu, et al, 2011;Xu et al, 2009). The LOD for the quantitative analyses were below 0.2 μg/kg, which is lower than the MRPL for AOZ residue set by the European Commission (European Commission., 2003).…”

“…Compared to other methods, the immunochromatographic test strip (ICTS) presents additional advantages: simplicity, speed, low cost and no requirement for skilled technicians or expensive equipment (Le et al, 2011;Le, Xie, Zhu, & Zhang, 2016). Recently, several reports on colloidal gold-based ICTS methods have been published for the detection of AMOZ (Li et al, 2015), SEM (Tang, Xu, Wang, Xiang, & Yang, 2011;Yin, Liu, Song, Kuang, & Xu, 2015) and AHD (Tang, Xu, Liu, et al, 2011;Xu et al, 2009), but there have been no reports of colloidal gold-based ICTS method being used to measure AOZ in animal tissues. Based on the monoclonal antibody (McAb) against AOZ's derivates (NPAOZ) produced in our lab (Le & Yu, 2015), the aim of this study was to develop a rapid ICTS using colloidal gold-antibody probe for quantitative and sensitive detection of AOZ in animal tissues.…”

“…Compared to other methods, ICA offers many advantages such as no requirement for instruments, simple operation, cheap price of detection and easy result evaluation. To date, for nitrofurans, there have been only three ICAs published for the detection of 1‐aminohydantoin (AHD, the metabolite of nitrofurantoin) and semicarbazide (SEM, the metabolite of nitrofurazone) . Nevertheless, in these three ICAs, the analyte is the derivative of AHD or SEM rather than the metabolite itself, e.g.…”

“…[7][8][9][10] But expensive equipment and well trained operators are pre-requisite for accurate detection. Furthermore, the tedious derivatisation and solid-phase extraction are still required for the analysis of marker residues in the rapid methods, including enzyme-linked immunosorbent based assay, [11][12][13][14] lateral ow immunoassay, 15 impedimetric immunoassay 16 and lateral ow biosensor assay. 17,18 The organic extraction solvent can be involved in the hybridization, which may interfere with the accuracy and reproducibility of the assay.…”
