Determination of histamine, 1-methylhistamine and N-methylhistamine by capillary electrophoresis with micelles

Abstract: Urinary histamine and Ngamma-methylhistamine (1-MH), a histamine metabolite, are highly correlated with histamine in plasma. Therefore, allergic reactions can be examined by determination of histamine and 1-MH in urine. We separated histamine, 1-MH and Nalpha-methylhistamine (N-MH) by capillary electrophoresis with UV detection at 210nm, using borate buffer (pH 9) containing 100 mM SDS. The absolute detection limits were 200, 100 and 50 pg for histamine, 1-MH and N-MH, respectively. To purify histamine 1-MH an… Show more

“…Serum levels of 1-methylhistamine are higher in asthmatic patients; they increase acutely after an asthma attack and are decreased by antiallergy medications. [42][43][44] Although histidine levels have been reported as increased in asthmatic patients compared with those in healthy control subjects, 34 they have not been reported in comparison with levels in patients with COPD.…”

“…They rise acutely after an asthma attack and are lowered by antiallergy medications. [42][43][44] Kynurenine is a product of tryptophan catabolism by the enzyme indoleamine 2,3, dioxygenase. 45 Indoleamine 2,3, dioxygenase activity is an important element in immune regulation in health and disease, including allergy.…”

“…Due to HA low concentrations in these different fluids even after an important release, sensitive quantification methods are needed. Various analytical methods have already been developed to identify and quantify HA in biological samples: liquid chromatography (LC) [9][10], gas chromatography (GC) [11][12], capillary electrophoresis (CE) [13] coupled with different detection modes. Derivatization was usually used to improve the sensitivity, but was time-consuming, and increased the risk of low analyte recovery.…”
