Pesticide residues in meat is of growing concern due to possible adverse effects on humans. Pesticide levels were assessed in five edible cattle parts: muscle, liver, kidney and tongue tissues to determine human health risk associated with consumption of these tissues. Health risk estimates were analysed using estimated daily intake (EDI), hazard quotient (HQ) and hazard index (HI) for two (2) age/weight categories: 1–11years/30 kg for children while 70 kg was used for adult. Risks were categorized for non-carcinogenic and carcinogenic health effects and measured at the average, maximum, 50th and 95th percentiles of the measured exposure concentrations (MEC). Total pesticide residues ranged from 2.38 to 3.86 μg/kg (muscle), 3.58 to 6.3 μg/kg (liver), 1.87 to 4.59 μg/kg (kidney) and 2.54 to 4.35 μg/kg (tongue). Residual pesticide concentrations in the tissues were in the order: Liver > Tongue > Muscle > Kidney. The concentrations of all the assessed pesticides observed in the tissues were however lower than the recommended maximum residual limits (MRLs). Human health risk estimations for the children showed EDI values for heptachlor epoxide, aldrin and dieldrin exceeding threshold values. Non-cancer risk posed to children on consumption of contaminated cattle parts showed HQ values for heptachlor epoxide, aldrin, dieldrin and HI values for organochlorines exceeding 1, indicating the possibility of non-carcinogenic health risks to consumers especially children from consumption of cattle meat from the selected abattoirs.

SummaryIn the present study three different cell cultures, derived from rabbit kidney (RK13), rat liver (WBF344) and insect origin (Sf21), were used to examine the cytotoxic effect of the insecticide endosulfan. Cytotoxicity was determined on the basis of cell proliferation activity; the cellular damage was assessed by evaluation of cytopathic effect and lactate dehydrogenase (LDH) leakage. Endosulfan treatment suppressed proliferative activity in cell cultures as follows: insect Sf21 cells (10 -2 M (P<0.05) miktarında belirlendi. Bu sonuçlar endosülfan maruziyetine karşı hücre tipine bağlı duyarlılığı işaret etmektedir. Endosülfan, memeli hücre kültürüne göre böcek kökenli hücre kültürlerindeki hücre sayılarında daha belirgin bir düşüşe neden olsa da LDH fazlalığı ve hücredeki mikroskobik hasar belirteçleri en belirgin olarak karaciğer hücrelerinde görülmektedir.

A rapid and effective method was developed for the analysis of 39 pesticide residues in porcine meat with dual-layer multiplug filtration cleanup and gas chromatographytandem mass spectrometry detection. The cleanup process was performed with columns packed with two layers, namely multiwalled carbon nanotubes, C18 and anhydrous magnesium sulfate (MgSO 4 ) as top layer, while mixture of florisil and MgSO 4 as bottom layer. A single-layer method was tested in parallel, with columns packed with the same amount of absorbents. Extraction conditions and filtration cleanup process were optimized to obtain satisfied method performance. Method linearity was calculated with coefficients of determination more than 0.995. The limits of quantitation were verified with acceptable accuracy at the lowest spiked concentration of 0.01 mg/kg (except pyrimethanil). The recoveries at three fortified levels (0.01, 0.05, and 0.1 mg/kg) in five replicates were between 74 and 118% (except pyrimethanil) with relative standard deviations range from 1 to 16%. The matrix effects were in the range of 1.01 to 2.84. This new method was applied for the analysis of multipesticide residues in market samples of porcine meat. This study showed the dual-layer multiplug filtration cleanup demonstrated better performance than that with the single-layer columns in cleanup of porcine meat.
