In present investigation, an attempt was made to develop gastro retentive tablets of cefpodoxime proxetil (CP) using locust bean gum as release retarded material. CP is an orally administered, extended spectrum, semi-synthetic antibiotic of cephalosporin class. CP has a short elimination half-life and also possesses high solubility, chemical, enzymatic stability and absorption profiles in acidic pH which makes CP suitable candidate for formulating it as gastro retentive dosage form for improved bioavailability. Sodium bicarbonate and citric acid were used as effervescent agents to get desired floating properties. The tablets prepared were evaluated and found to have acceptable physicochemical properties. The in vitro release data of optimized formulation was treated with mathematical equations and was concluded that drug release followed zero order kinetics with anomalous transport mechanism. Based on the results it can be concluded that floating tablets of cefpodoxime proxetil containing locust bean gum provides a better option for controlled release action and improved bioavailability.

The aim of this work was to study the influence of process variables on the quality attributes of pomegranate extract loaded solid lipid nanoparticles (PE-SLNs) using Plackett–Burman design. PE-SLN formulations were prepared by hot homogenization followed by ultra-sonication technique and evaluated based on the dependent variables that were analyzed utilizing Statgraphics Centurion XV software. The lipid and surfactant (type and concentration), co-surfactant concentration, sonication time, and amplitude were selected as the independent variables ( X 1 – X 7 ). The dependent parameters were particle size, polydispersity index, zeta potential, entrapment efficiency, and cumulative drug release ( Y 1 – Y 5 ). Response surface plots, Pareto charts, and mathematical equations were generated to study the influence of independent variables on the dependent quality parameters. Out of seven variables, X 1 , X 2 , and X 6 have the main significant ( p value < 0.05) effect on the entrapment efficiency, the cumulative drug release, the polydispersity index, respectively, while particle size was mainly affected by X 3 , X 6 and zeta potential by X 1 , X 3 , and X 4 . Consequently, this screening study revealed that stearic acid as lipid, Tween 80 as surfactant, as well as sonication with short time and high amplitude can be selected for the development of PE-SLN formulation with minimum particle size, maximum zeta potential, highest entrapment, and sustained drug release behavior. Meanwhile, concentrations of lipid, surfactant, and co-surfactant are planned to be scaled up for further optimization study. In conclusion, the Plackett–Burman design verified its influence and significance in determining and understanding both process and formulation variables affecting the quality of PE-SLNs.

Migraine has long been regarded as a vascular disorder because of the throbbing nature of the pain. Most patients with migraine require pharmacologic treatment. In the present research work sumatriptan succinate is used in the form of compressed core tablets via oral route of administration for effective treatment of migraine. The aim of this study was to reduce the dosing frequency and avoid hepatic first pass metabolism by preparing sumatriptan succinate loaded alginate microsphere. The microspheres were prepared by emulsification method. The prepared microsphere were characterized by scanning electron microscopy, and evaluated for different parameters like particle size, entrapment efficiency, polydispersity index, surface charge and in vitro drug release. The microsphere loaded compressed core tablets were prepared by direct compression method, where the drug loaded microsphere was present in the core of tablet. Further the outer coating layer was applied on the core of tablet that contains plain sumatriptan succinate to immediate release and provides instant relief from migraine symptoms. The formulations were evaluated for various parameters as well as in vitro drug release and compared with plain sumatriptan succinate loaded compressed core tablet. The in vitro drug release showed immediate drug release within 2 min from outer coating layer as well as sustained drug release for up to 24 h from core of tablet. It is concluded that the formulation provide instant as well as delayed release of drug to migraine patient which can decrease the dosing frequency and increase patient compliance.
