Cytotoxicity of tamoxifen in normal and tumoral cell lines and its ability to induce cellular transformation in vitro

Abstract: Tamoxifen (TAM) is a non-steroidal anti-estrogen used to treat patients with estrogen receptor-positive breast cancer and as a chemopreventive agent against breast cancer in high risk pre- and post-menopausal women. However, recent studies have shown that tamoxifen causes endometrial and hepatic cancer. In this study, we examined the effects of tamoxifen (5, 10, 25 and 50 microM) on the growth and proliferation of nine tumoral cell lines (UACC62, MCF-7, NCI-460, K-562, OVCAR-03, PC-03, HT-29, 786-0, NCI-ADR) a… Show more

“…According to the latest report from the International Agency for Research on Cancer/WHO, the incidence was estimated at 12.7 million new cases annually, with cancer causing the deaths of 7.6 million people worldwide per year 8 . Tamoxifen (TAM) is a synthetic no steroidal antiestrogen, it is commonly prescribed to treat patients with breast cancer 9 . TAM belongs to the family of selective modulators of estrogen receptors.…”

“…Tamoxifen may be having same effect in colon carcinoma. Tamoxifen also demonstrates its effect through growth factors and immune system, and thus, it provides certain benefits for estrogen-neutral patients (11)(12)(13). The aim of our study is to investigate the effects of quercetin and tamoxifen on apoptosis in CaCo-2 human colon adenocarcinoma cell line.…”

“…Quercetin can be also modulate estrogen receptor activation (6)(7)(8)(9)(10). Another agent we used in our study, tamoxifen is a non-steroidal anti-estrogen drug commonly used in the treatment of estrogen receptor positive breast cancer (11,12). Among the effect mechanisms of tamoxifen is to bind to estrogen receptor in order to prevent the recurrence of breast carcinoma and to inhibit the growth of cancer cells.…”
