A strategy to control the black sigatoka disease (Mycosphaerella fijiensis Morelet) of plantain (Musa ssp ., AAB group) in Africa, targeting the incorporation of durable host plant resistance, was initiated at the International Institute of Tropical Agriculture (IITA) . The commonly accepted intractability of plantain to genetic improvement has been challenged by the identification of 37 different, seed-fertile plantain cultivars and by the production of 250 hybrids in four years of breeding work . Twenty tetraploid hybrids have been selected for their increased black sigatoka resistance, high yields, large parthenocarpic fruits and improved ratooning. `Calcutta 4' (Musa acuminata spp . burmannicoides) was the diploid male parent of 17 of the selected hybrids, which indicates that the inferior bunch characteristics of this wild banana were generally not transmitted to its tetraploid progenies . Conversely, the 4x progeny of plantain readily expressed black sigatoka resistance when crossed with `Calcutta 4' . Progenies of the triploid plantain cvs . 'Obino l'Ewai' and Bobby Tannap' differed in their black sigatoka breeding values, the former producing larger numbers of promising hybrids . Tetraploids obtained from crosses of plantain cultivars with the homozygous `Calcutta 4' displayed variation in black sigatoka reaction, qualitative morphological traits and growth and yield parameters, suggesting the occurrence of segregation and recombination during the modified megasporogenesis leading to the formation of 2n eggs in the triploid female plantain parents . The variation in black sigatoka reaction among the tetraploid progenies of plantain suggests that resistance could be regulated by recessive, additive genes .

Polyploidy was induced in two Musa species and their hybrid by means of immersing newly germinated diploid seedlings in 0.5% aq. colchicine solution. Tetraploid M. balbisiana plants were taller and more robust but had slower growth rate, droopier leaves, fewer suckers, and scantier root systems than the diploids. Tetraploidy affected fruit size and shape in M. balbisiana and in M. acuminata. Tetraploidy did not affect bunch size (the number of fruit‐bearing nodes) in M. acuminata banksii but caused great reduction in bunch size of M. acuminata microcarpa ‘Zebrina.’ Doubling of the chromosome number resulted in doubling of the anthocyanin concentration in the leaves of the pigmented banana plants. Colchicine‐induced female sterility was detected in a treated diploid plant of M. acuminata subsp. microcarpa. A number of either female or male, or both female and male sterile plants were encountered in colchicine‐induced tetraploid M. acuminata species. Only one secondary M. balbisiana tetraploid plant was found to be partially female sterile. Colchicine treatment of the seedlings resulted in chromosomal irregularities at mitosis. Chromosomes appeared as open chains, rings and multiple configurations at metaphase. In the majority of cases doubled chromsomal complements reverted to their original number. However, there were cases in which diploids were induced to tetraploidy and later reverted to triploidy instead of diploidy. Also, there were cases in which triploids were doubled to hexaploidy and later reverted to tetraploidy instead of triploidy. From the close morphological similarity between a number of triploid and tetraploid M. balbisiana plants and some edible triploid cultigens belonging to the “ABB Group,” it is suggested that the karyotype of some of these cultigens could be “BBB” rather than “ABB” which contains one “A” M. acuminata genome. New pathways and additional evolutionary possibilities are incorporated into Simmonds’ scheme on the evolution of edible bananas.

Breeding of banana is hampered by its genetic complexity, structural chromosome rearrangements and different ploidy levels. Various scientific disciplines, including cytogenetics, linkage mapping, and bioinformatics, are helpful tools in characterising cultivars and wild relatives used in crossing programs. Chromosome analysis still plays a pivotal role in studying hybrid sterility and structural and numerical variants. In this study, we describe the optimisation of the chromosome spreading protocol of pollen mother cells focusing on the effects of standard fixation methods, duration of the pectolytic enzyme treatment and advantages of fluorescence microscopy of DAPI stained cell spreads. We demonstrate the benefits of this protocol on meiotic features of five wild diploid Musa acuminata bananas and a diploid (AA) cultivar banana “Rejang”, with particular attention on pairing configurations and chromosome transmission that may be indicative for translocations and inversions. Pollen slides demonstrate regular-shaped spores except “Rejang”, which shows fertile pollen grains of different size and sterile pollen grains, suggesting partial sterility and unreduced gamete formation that likely resulted from restitutional meiotic divisions.
