Cytogenetic studies in peripheral blood of bovines afflicted by papillomatosis

Abstract: Ten types of bovine papillomavirus (BPV) have been described and there are reports of viral transmission via blood. The presence of viral DNA in lymphocytes was described to be associated with chromosome instability in these cells. This study presents an evaluation of chromosome instability in short-term peripheral lymphocyte cultures from cows presenting skin papillomatosis, compared with asymptomatic infected animals and non-infected healthy bovines. In a total of 2203 cells, 918 (42%) showed at least one ch… Show more

“…The comet assay showed a high clastogenic level in PBMCs infected with BPV. This result reinforces the point that DNA breaks result from viral activity, leading to genomic instability and eventual malignancy (Stocco dos Santos et al, 1998;Melo et al, 2011;Araldi et al, 2013a). The clastogenic action of HPV has already been reported and is associated with spontaneous abortions (Calinisan et al, 2002).…”

“…The presence of BPV sequences in peripheral blood has already been described in the literature (Stocco dos Santos et al, 1998;Roperto et al, 2008;Diniz et al, 2009;Melo et al, 2011;Araldi et al, 2013b). The peripheral blood does not represent the first site of virus contact, according to the infection pathway commonly described (Florin et al, 2012).…”

“…The presence of BPV-2 in peripheral blood leads to chromatin changes (Stocco dos Santos et al, 1998;Melo et al, 2011;Araldi et al, 2013), which result in genomic instability, evidenced by chromosomal loss and gain. This event is frequently observed in solid tumors, with it being associated with the oncogenic process (Duensing and Münger, 2002;Toledo, 2011;Córtes-Gutiérrez et al, 2012).…”

“…Considering specific situations in which clinical signs that suggest malignance could not be observed, BPV infection could cause cytogenetic aberrations, leading to genomic instability and cancer (Stocco dos Santos et al, 1998;Melo et al, 2011). The bovine lifetime is long enough to allow cancer to arise from years of accumulated mutations.…”
