Culture-based identification of pigmented Porphyromonas and Prevotella species in primary endodontic infections

Abstract: Background. The most common species isolated from primary endodontic infections are black-pigmented bacteria. These species are implicated in apical abscess formation due to their proteolytic activity and are fastidious in nature. Therefore, the present study was carried out to evaluate the presence and identification of various pigmented Porphyromonas and Prevotella species in the infected root canal through culture-based techniques.Methods. Thirty-one patients with primary endodontic infections were selected… Show more

“…In our study, samples were obtained from the single largest necrotic root canal to narrow down the microbial growth evaluation to a single ecological condition 10,11 . To determine the antimicrobial effectiveness of any remaining irrigant in the canals after obtaining initial samples, the pulp chambers were sealed with cotton and Glass Ionomer cement for next 48 hours.…”

“…The broth culture will look cloudy after incubation in the incubator for 48 hours. In order to recommended for culture, P gingivalis is the PRAS Brucella Blood Agar Plate from the anaerobic system (AS-111) 12,14,15 To test the antibacterial activity of mouthwash preparations, there are 9 paper disks were prepared, each 3-paper disk was dipped in a) each mouthwash formula, b) standard solution infusion to the combination of betel leaf, bay leaf, and c) negative control (that was additives without the active of infusion were bay leaf and betel leaf) d) positive control (chlorhexidine). Solutions a), b), c) and d) were provided as much as 20 mL each.…”

“…Black-pigmented organisms such as Prevotella followed by Porphyromonas , especially, P. gingivalis are the most isolated microorganisms through culture of samples obtained from necrotic pulps. [ 34 ] While Fusobacterium , Parvimonas , and Peptostreptococcus were indicated as the most prevalent microorganisms in acute endodontic infections, phyla Firmicutes , Bacteroidetes , and Actinobacteria were the most prevalent microorganisms in chronic endodontic infections.
