Cryopreservation of winter-dormant apple buds: establishment of a duplicate collection of Malus germplasm

Abstract: The clonal wild-species Malus collection at the Julius Kühn-Institute, Institute for Breeding Research on Fruit Crops, Germany, comprises 507 accessions of 46 species and represents one of the largest collections in Europe. Cryopreservation was applied to 77 accessions of 31 species and hybrids in the Malus collection using a winter vegetative-bud method. No significant differences were observed in the recovery of the two controls, untreated buds and desiccated buds, however, cryopreservation significantly dec… Show more

“…This assumption is supported by results of a previous study with Malus, as the buds with the smallest volumes had the highest survival of the primary buds when grafted following cryopreservation . Moreover, the recovery of Malus buds can be attributable to a secondary bud outgrowth Höfer 2015). In the case of cv.…”

“…This protocol is generally used with Malus (e.g. Volk et al 2008;Grout et al 2011;Lambardi et al 2011;Höfer 2015;Höfer and Hanke 2017) and also applied for Salix (Towill and Widrlechner 2004;Jenderek et al 2014) Fraxinus (Volk et al 2009) Juglans, Prunus, (Jenderek et al 2014) and Vaccinium . Protocols where recovery of buds is done via in vitro culture have been developed e.g.…”

“…Additionally, pests and diseases can threaten the material, especially the less adapted or susceptible accessions. One of the important apple diseases, fire blight, caused by the bacterium Erwinia amylovora, can severely damage or even eradicate susceptible apple accession [33,34]. In 2013, the Allium field collection at IPK was challenged with a serious infestation of larval stage click beetles (Elateridae), also known as wireworms.…”

“…Криоконсервация спящих почек используется в основном для древесных плодовых культур умеренного климата, в частности, яблони (Forsline et al, 1999;Towill et al, 2004;Volk et al, 2008;Lambardi et al, 2009;Höfer, 2015). Однако для древесных тропических культур данный метод не подходит.…”
