Recently, many nutraceutical products containing the powdered or extracted parts of C. militaris have become available for health care. Due to the increased morbidity and mortality, poisonings associated with the use of herbs have raised the universal attention. Herein, we carried out the 28-day repeated toxicity test in male and female ablactated rats (three weeks old) given C. militaris powder orally at 0 (control), 1, 2, and 3 g/kg per day. Noticeable increments of serum aspartate and alanine aminotransferase (ALT and AST) levels were observed for both sexes, suggestive of weak hepatic toxicity. Nephrotoxicity characterized by tubular epithelium degeneration and necrosis was observed at the high dose, and the male rats were more susceptible to renal toxicity than female rats. In addition, the genes and protein expressions of novel markers of kidney toxicity, such as kidney injury molecule-1 (KIM-1) were enlarged in the renal cortex and the urine. Moreover, C. militaris treatment significantly decreased superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activities. However, the ratio of glutathione oxidized form (GSSG)/glutathione reduced form (GSH) was increased by C. militaris treatment. We conclude that dietary contamination with C. militaris may have renal toxicity potentials, at least in part by causing oxidative damage to the kidney.

Authentication and quality assessment of Cordyceps sinensis, a precious and pricey natural product that offers a variety of health benefits is highly significant. To identify effective chemical markers, authentic C. sinensis was thoroughly screened by using HPLC-MS/MS. In addition to many previously reported ingredients, two glycosides, i.e. cyclo-Ala-Leu-rhamnose and Phe-o-glucose were detected for the first time in this material. Six ingredients detected, including cordycepin, D-mannitol, Phe, Phe-o-glucose, cyclo-Gly-Pro, and cyclo-Ala-Leu-rhamnose, were selected as a collection of chemical markers. An HPLC-MS/MS method was developed to simultaneously quantify them with sensitivity and specificity. The method had limits of detection ranging from 0.008 μg mL−1 for cordycepin to 0.75 μg mL−1 for cyclo-Gly-Pro. Recovery was found between 96% and 103% in all tests. To evaluate the effectiveness of the marker collection proposed, 5 authentic C. sinensis samples and 5 samples of its substitutes were analyzed. Cordycepin, D-mannitol, and Phe were found present in all samples. The contents ranged from 0.0076 to 0.029% (w/w) for cordycepin, 0.33 to 18.9 % for mannitol, and 0.0013 to 0.642% for Phe. Interestingly, the two glycosides, Phe-o-glucose and cyclo-Ala-Leu-rhamnose were detected only in authentic C. sinensis samples. These results indicated that the proposed protocol based on HPLC-MS/MS quantification of the markers might have a great potential in authentication and quality assessment of C. sinensis.

Cordyceps sinensis (Berk.) Sacc. ve Farmakolojik Açıdan Önemi Ö ÖZ ZE ET T Cordyceps sinensis, Uzak Doğu'da özellikle Çin, Nepal ve Tibet'in yüksek platolarında yetişen çok değerli ve nadir bulunan tıbbi bir mantardır. Çin'de MS 620'ye kadar uzanan yazılı literatürde; büyülü bir yaratık, hayvan ve bitki olarak tanımlanan tür, Geleneksel Çin Tıbbında yüzyıllardan beri ilaç olarak kullanılmaktadır. Caterpillar fungus, Dong Chong Xia Cao, Çin tırtıl mantarı veya Yarsagumba olarak da bilinen Cordyceps sinensis, taksonomik olarak Ascomycota (keseli mantarlar) sınıfı, Clavicipitaceae familyasında yer alan entomopatojen bir canlı türüdür. Meyveli gövdenin morfolojik özellikleri; hafif silindirik, çubuk şeklinde, renk olarak da sarımsı-turuncu veya kırmızımsı-turuncu renge sahip olarak tanımlanmaktadır. Bu mantar geleneksel olarak ishal, baş ağrısı, öksürük, romatizma, karaciğer ve böbrek hastalıkları, diyabet, yorgunluk gibi hastalıklar için binlerce yıldan beri kullanılmaktadır. Kimyasal içeriğinde bulunan nükleozitler, polisakkaritler, steroller, proteinler, vitaminler, mineraller sayesinde tıbbi olarak antitümör, immünomodülatör, antioksidan, antidiyabetik, cinsel gücü artırıcı, yorgunluk giderici, böbrek ve karaciğer fonksiyonlarını iyileştirici etkiler göstermesinin yanı sıra besin değeri olarak da önemli bir tıbbi mantardır. Kimyasal içeriğine bakıldığında, sahip olduğu nükleozit grubuna ait olan kordisepin ve kordisepik asit başlıca bileşenler olarak öne çıkmaktadır. Cordyceps sinensis, son yıllarda tüm dünyada popüler doğal ürünler arasında yerini almıştır. Özellikle antioksidan ve immünomodülatör etkileri nedeniyle, antitümör olarak kullanımı göze çarpmaktadır. Bu nedenle bu çalışmada, önemli farmakolojik etkilere sahip olan ve tıbbi olarak da kullanılan Cordyceps sinensis hakkında daha detaylı bilgi verilmesi, üzerine yapılmış olan araştırmaların sunulması amaçlanmıştır.
