Cord blood‐derived human cultured mast cells produce transforming growth factor β1

Abstract: Cord blood-derived human cultured mast cells constitutively express mRNA for TGF-beta1 and produce functional TGF-beta1. Because TGF-beta1 has been shown to be highly fibrogenic, these results may highlight a novel role for human mast cells in tissue fibrosis.

The real-time RT-PCR data also indicate that TGF-b1 is probably the main form of TGF-b produced by human MCs compared with TGF-b2.…”

“…Kanbe and colleagues (35) previously reported that human MCs produce TGF-b1. However, the concentration of TGF-b1 was too low to be detectable by ELISA in their experiments, and, to our knowledge, there are no other reports of the production of active TGF-b1 in human MCs other than reports of immunohistochemical staining of TGF-b1 in MCs (36).…”

“…In addition, IL-4-positive mast cells are signiWcantly associated with the size of allergic skin prick-test wheal reaction and serum total IgE levels [156]. Even though human mast cells have been shown to express IL-6 and TGF-1 [88,99]; it is, however, not known to what extent mast cells contain these Th17 priming cytokines in the psoriatic skin. Our preliminary results suggest that mast cells contain IL-6 immunoreactivity in the developing psoriatic lesion induced by the tape-stripping technique (Suttle et al, unpublished results) (Fig.…”

“…The mast cells produce a variety of fibrogenic cytokines, including plateletderived growth factor, transforming growth factor-b (TGF-b), and basic fibroblast growth factor. [3][4][5] These cytokines are involved in numerous inflammatory and neoplastic conditions in which marrow fibrosis is encountered and are thus not specific to mast cells. In addition, mast cell tryptase, which has been shown to be a powerful fibroblast mitogen, 6 is thought to play an important fibrogenic role in systemic mastocytosis.…”
