Constituents of the aerial parts of Eclipta prostrata and their cytotoxicity on human ovarian cancer cells in vitro

Abstract: A new terthiophene, 3'-hydroxy-2,2':5',2″-terthiophene-3'-O-β-D-glucopyranoside (1) and a new oleanane-type saponin, echinocystic acid-3-O-(6-O-acetyl)-β-D-glucopyranoside (7) were isolated from the aerial parts of Eclipta prostrata L. Moreover, five thiophenes (2-6), seven triterpenoids (8-14), two coumestans (15 and 16), and four flavonoids (17-20) having previously known chemical structures were isolated during the same course of this study. All the isolates 1-20 were evaluated for their cytotoxicity agains… Show more

“…The molecular mechanisms of the GCs-mediated induction of apoptosis involve down regulation or inhibition of anti-apoptotic molecules, such as Bcl-2, as well as upregulation or activation of pro-apoptotic molecules such as BAX-2 (Lotem and Sachs, 1995;Salomons et al, 1997;Rieger et al, 1999;Murphy et al, 2000;Das et al, 2004;Almawi et al, 2004;Hoijman et al, 2004;Mahmoud et al, 2009;Lopez-Royuela et al, 2010;Melarangi et al, 2012;Zaman et al, 2014). The additive effect of 10 μΜ EAs and 1 μM DEX on the NF-κΒ transcriptional inactivation and the induction of apoptosis at concentration lower than that reported in the literature may have an important role in immunosuppressive and anticancer combinations therapy, which aims at minimizing side effects of any single drug.…”

“…Also, an orally administered methanolic leaf extract exerted encouraging anticancer activity against Ehrlich ascites carcinoma in Swiss albino mice [89], and a hydroalcoholic extract reversed multidrug resistance in an animal model of liver cancer induced by diethylnitrosamine and 2-acetylaminofluorene [86]. Furthermore, terthiophenes, thiophenes, saponins, triterpenoids, coumestans, and flavonoids isolated from the aerial parts exhibited cytotoxicity against cultured SKOV3 human ovarian cancer cells [90]; an eclalbasaponin I-containing fraction from the aerial parts and the saponin dasyscyphin-C isolated from the leaves inhibited the in vitro proliferation of SMMC-7721 human hepatocarcinoma and HeLa human cervical carcinoma cells, respectively [85,91]; and eclalbasaponin II induced cytotoxicity as well as apoptotic and autophagic cell death in human ovarian cancer cell lines [92].…”

“…In synopsis, our outcomes show that Eclipta alba has colossal potential in reciprocal and elective drug for the treatment of malignant growth. [7] Kim HY et al have been reported another terthiophene, 3'-hydroxy-2,2':5',2″-terthiophene-3'-O-β-Dglucopyranoside (1) and another oleanane-type saponin, echinocystic corrosive 3-O-(6-O-acetyl)-β-Dglucopyranoside (7) were disconnected from the flying parts of Eclipta prostrata L. Additionally, five thiophenes (2-6), seven triterpenoids (8)(9)(10)(11)(12)(13)(14), two coumestans (15 and 16), and four flavonoids (17)(18)(19)(20) having recently realized synthetic structures were secluded amid a similar course of this examination. All the disconnects 1-20 were assessed for their cytotoxicity against human ovarian disease cells (SKOV3) utilizing MTT measures.…”
