Complete genome sequence of a novel porcine parainfluenza virus 5 isolate in Korea

Abstract: A novel cytopathogenic paramyxovirus was isolated from a lung sample from a piglet, using continuous porcine alveolar macrophage cells. Morphologic and genetic studies indicated that this porcine virus (pPIV5) belongs to the species Parainfluenza 5 in the family Paramyxoviridae. We attempted to determine the complete nucleotide sequence of the first Korean pPIV5 isolate, designated KNU-11. The full-length genome of KNU-11 was found to be 15,246 nucleotides in length and consist of seven nonoverlapping genes (3… Show more

“…Moreover, the UAA termination codon is also mutated, appearing as CAG in strain CPI + /CPI − and as CAA in strain SER/KNU-11. The hypothesis that SH is not expressed by these strains, as was adduced in relation to strain KNU-11 (22), was tested by immunoblotting of lysates of Vero cells infected with strain W3, strain CPI + , variant strain CPI − , or strain SER using a polyclonal peptide antiserum raised against the N-terminal 16 residues of strain W3 SH. The sequence of this region is well conserved among strains (Fig.…”

“…Many new paramyxoviruses have been identified recently (11)(12)(13)(14)(15). Isolating infectious viruses and studying them in suitable animal models will enable us to understand their pathogeneses.…”

“…In addition, the complete genome of a PEDV strain, KNU-1305, was sequenced and analyzed. The 5′ and 3′ ends of the KNU-1305 genome were determined by rapid amplification of cDNA ends as described elsewhere ( 11 ). Ten overlapping cDNA fragments were generated to encompass the entire genome, pooled in equimolar amounts, and subjected to next-generation sequencing as previously described ( 12 ); the sequencing reads were assembled by using complete PEDV reference genomes from GenBank ( 13 , 14 ).…”
