Comparison of the Troponin T and Troponin I ELISA Tests, as Measured by Microplate Immunoassay Techniques, in Diagnosing Acute Myocardial Infarction

Abstract: Summary:We describe an improved procedure using a standard microplate immunoassay reader to measure the concentration of troponin T in human serum. We also describe an immunoassay for troponin I in serum. Only 160 μΐ of serum are needed for a single analysis of each troponin. For comparison, creatine kinase MB mass analysis in serum was performed with a commercial luminometric method. From 95 apparently healthy people the following values were obtained: creatine kinase MB mass 2.6 ±1.2 μg/l, troponin Τ 0.027 ±… Show more

“…The Cloud-Clone Corp. Rat Troponin I ELISA kit (Cloud-Clone Crop., Houston, USA) was employed to conduct a quantitative sandwich immunoassay for determining the level of troponin I in the serum in keeping with the approach suggested by Penttilä et al [ 30 ]. Meanwhile, the level of creatine kinase-MB enzyme in the serum (CK-MB) was determined with the assay kit supplied by Bioassay Systems (Hayward, CA, USA) and based on the method by Bishop et al [ 31 ].…”

“…The CK activity was analyzed according to Szasz et al, [ 19 ] whereas serum CK‐MB activity was analyzed according to Würzburg et al [ 20 ] Serum IL‐2, IL‐6, and TNF‐α were measured using ELISA kits from R&D, following the manufacturer's instructions. Serum troponin I levels were also determined using an ELISA kit (Spectrum Diagnostics), as described by Penttilä et al [ 21 ]…”

“…In this context, different immunological methodologies have been developed to detect hcTnI in human samples. Electrochemiluminiscent immunoassays (ECLIA) [ 14 , 15 , 16 ], Fluoroimmunoassays [ 16 , 17 , 18 , 19 , 20 ], and enzyme-linked immunosorbent assay (ELISA) [ 21 , 22 , 23 , 24 ] have been widely used for AMI diagnosis. However, these techniques require expensive laboratory equipment, well-trained personnel and are time-consuming techniques, especially taking into account that a timely and reliable diagnosis is required for appropriate patient treatment.…”
