Comparison of platelet quality and function across apheresis collection platforms

Abstract: Background Platelet concentrates (PLT) can be manufactured using a combination of apheresis collection devices and suspension media (plasma or platelet additive solution (PAS)). It is unclear how platelet quality and hemostatic function differ across the current in‐use manufacturing methods in the United States. The objective of this study was therefore to compare baseline function of PLT collected using different apheresis collection platforms and storage media. Study Design and Methods PLT were collected at … Show more

“…Collectively, these studies show that there are significant differences in the levels of hemolysis, potassium, cytokine and microparticle levels, oxidative stress, oxygen carrying capacity, deformability, and residual plasma, platelet and leukocyte concentrations across the different methods used to manufacture an RBC product. Similar studies have been performed for platelet [ 34 , 35 ] and plasma [ 36 ] components highlighting the variability that exists in “what is in the bag”.…”

“…Platelet activation is known to occur during component preparation (in both apheresis and WB‐derived platelets) or when platelets are stored for extended durations at both room and cold temperatures 19,22,23 . In our study, storage of Reveos PCs at room temperature for 7 days resulted in higher P‐selectin expression compared to day 1, a trend that has been observed in PCs produced with Reveos and by manual method 3,24 .…”

“…Platelet aggregation response of stored platelets to single and dual agonists was evaluated as described previously 19 . Light transmission aggregometry (Chrono‐log Model 700 aggregometer, Chrono‐log Corp, Havertown, PA) was performed with stored platelet samples (250 × 10 3 /μL, diluted with PPP) and the agonists adenosine diphosphate (ADP; 5 μM), collagen (1 μg/mL), epinephrine (2 μM), ADP (5 μM) + collagen (1 μg/mL), and ADP (5 μM) + epinephrine (2 μM).…”
