To understand the fundamental mechanisms behind malignancy of breast tumors and also contribute to the discovery of improved methods for prevention, diagnosis and treatment, animal cancer models remain essential. We aimed to establish an optimal orthotopic cancer model for breast cancer in the immunocompetent Swiss mouse, describing the detailed microanatomy of the mammary glands, the sentinel lymph node and lymphatic mapping, evaluating histopathological changes and characterizing the tumor by computed microtomography and interleukins expression. The inoculation of fresh Ehrlich tumor cells led to a detectable tumor as early as 24 hours later; after 7 days, mammary, muscular, dermal vascular and lymphatic invasion were observed and also micrometastases in mammary adipose tissue, sentinel lymph node and contralateral lymph node. From the inoculation site the tumor invaded the host mammary gland structures, the dermis and endomysium of skeletal muscle tissue. Type 1 T helper cytokine levels (IL-1β and IL-17) were signifi cantly higher than anti-infl ammatory Type 2 T helper (IL-4) after inoculation of fresh tumor cells. Differently, frozen tumor cells induced tumor development only 14 days after the inoculation while presenting expression of distinct interleukins. The set of fi ndings indicates that orthotopic transplantation provides the microenvironment critical for cell interactions involved in the development of cancer and subsequent metastasis. In this regard, the anatomical and physiological knowledge of the mammary glands and ductal networks favor studies, diagnoses and therapies related to breast cancer and metastasis. Moreover, this study provides anatomical support for the understanding of the lymphatic spread process of cancer cells.

ABSTRACT:This study was to evaluate, the pro-apoptotic activity of ethanolic Chelidonium majus L. (CM) leaf extract and/or gamma-radiation in Ehrlich ascites carcinoma (EAC) bearing mice by measuring tumor volume, apoptotic factors (caspase-3, Bcl-2 and Bax proteins, tumor necrosis factor-alpha (TNF-α), angiogenesis factors (matrix metalloproteinase (MMP-2 and MMP-9); tissue inhibitor of metalloproteinases (TIMP-1), DNA fragmentation, besides histopathological examination. After tumor inoculation, mice received CM (100 mg/body weight/day) on the 10 th day and were exposed to whole body γ-radiation (6Gy) on the 17 th day. The data obtained reveales that combining CM with γ-radiation exposure induce significant regression in tumor growth, up-regulate Bax, caspase-3, TIMP-1 and inhibit Bcl-2, TNF-α, MMP-(2 and 9). Cytotoxicity is substantiated by increased DNA-fragmentation (comet assay and DNA content) and histological examination. It could be suggested that combining CM with gamma-radiation increased apoptosis of tumor cells which would help to increase the lifespan of mice.

The present study aimed to investigate the mode of action of nano-CaPs in vivo as a therapy for solid tumor in mice. To achieve this goal, Ehrlich Ascites Carcinoma (EAC) was transplanted into 85 Swiss male albino mice. After nine days, the mice were divided into 9 groups. Groups 1 and 2 were allocated as the EAC control. Groups 3 and 4 were injected once intratumorally (IT) by nano-calcium phosphate (nano-CaP). Groups 5 and 6 received once intraperitoneal injection (IP) of nano-CaP. Groups 7, 8, and 9 received nano-CaP (IP) weekly. Blood samples and thigh skeletal muscle were collected after three weeks from groups 1, 3, 5, and 7 and after four weeks from groups 2, 4, 6, and 8. On the other hand, group 9 received nano-CaP (IP) for four weeks and lasted for three months to follow up the recurrence of tumor and to ensure the safety of muscle by histopathological analysis. Tumor growth was monitored twice a week throughout the experiment. DNA fragmentation of tumor cells was evaluated. In thigh tissue, noradrenaline, dopamine, serotonin (5HT), and gamma-aminobutyric acid (GABA) were measured. In serum, 8-Hydroxy-deoxyguanosine (8-OHDG), adenosine triphosphate (ATP), and vascular endothelial growth factor (VEGF) were analyzed. Histopathological and biochemical results showed a significant therapeutic effect of nano-CaP on implanted solid tumor and this effect was more pronounced in the animals treated IP for four weeks. This improvement was evident from the repair of fragmented DNA, the significant decrease of caspase-3, 8-OHDG, myosin, and VEGF, and the significant increase of neurotransmitters (NA, DA, 5HT, and GABA). Additionally, histopathological examination showed complete recovery of cancer cells in the thigh muscle after three months.
