Collagen Dynamics During the Process of Osteocyte Embedding and Mineralization

Abstract: Bone formation, remodeling and repair are dynamic processes, involving cell migration, ECM assembly, osteocyte embedding, and bone resorption. Using live-cell imaging, we previously showed that osteoblast assembly of the ECM proteins fibronectin and collagen is highly dynamic and is integrated with cell motility. Additionally, osteoblast-to-osteocyte transition involved arrest of cell motility, followed by dendrite extension and retraction that may regulate positioning of embedding osteocytes. To further under… Show more

“…For instance, (Qiu et al, 2020) has fabricated an injectable periosteal ECM hydrogel that dynamically integrates multiple biological functions, promotes angiogenesis and osteogenesis at the defect site. The whole dynamic process contains bone formation, remodeling and repair, which involves cell migration, ECM assembly, osteocyte embedding, and bone resorption (Shiflett et al, 2019). Our experimental results have proved the effect on cell growth, migration, and differentiation.…”

“…Recently, it is demonstrated that the process by which osteocytes push type I collagen fibers outward from the center of the formed lacuna mediates osteocytes lacunae formation, which is accompanied by increased collagen deposition and collagen-fiber network compaction surround the lacunae. Therefore, the dynamic assembly of bone collagen contributes greatly to the encapsulation and mineralization of osteocytes in bone matrix (Shiflett et al, 2019).…”

“…Osteocytes are the longest-lived bone cells, accounting for 90-95% of all cells in mineralized bone. In contrast, osteoclasts and osteoblasts make up approximately 5% of bone tissue cells (Shiflett et al, 2019). Osteocytes embedded in lacuna within the mineralized matrix form dendritic processes within canaliculi to communicate with neighboring osteocytes and other cell lineages (Zhang et al, 2012).…”
