Mali et al.: Carboxymethyl cellulose-based hydrogel films for drug deliveryThe objective of this study was to synthesize and characterize citric acid crosslinked hydrogel films of carboxymethyl cellulose-tamarind gum for topical drug delivery. The hydrogel films were characterized by attenuated total reflectance-Fourier-transform infrared spectroscopy, solid-state 13 C-nuclear magnetic resonance spectroscopy and differential scanning calorimeter. The prepared hydrogel films were evaluated for the carboxyl content and equilibrium swelling ratio. Moxifloxacin hydrochloride was loaded into these hydrogel films and drug release was monitored in the phosphate buffer pH 7.4. Haemolysis assay was used to study biocompatibility of hydrogel films. Results of the attenuated total reflectance-Fourier-transform infrared spectroscopy, solid-state 13 C-nuclear magnetic resonance and differential scanning calorimeter confirmed the formation of citric acid-crosslinked hydrogel films. Total carboxyl content of hydrogel film was found to be increased when polymer ratio and amount of citric acid was increased. In contrast, swelling of hydrogel film was found to be decreased with increase in polymer ratio and amount of citric acid. Batch B1 showed highest drug loading with non-Fickian release mechanism. All remaining batches showed nonFickian release behavior with diffusion coefficient greater than 0.5. Results of haemolysis assay indicated that the citric acid crosslinked carboxymethyl cellulose-tamarind gum hydrogels were safe to be used in drug delivery. These results indicated that the citric acid crosslinked carboxymethyl cellulose-tamarind gum composite hydrogel film has the potential to be used in topical novel drug delivery systems.

β‐cyclodextrin grafted hydroxyethylcellulose (βCD‐g‐HEC) hydrogel films were prepared for the controlled release of poorly soluble model drug (ketoconazole) using citric acid as crosslinking agent. The active βCD and carboxyl content of the hydrogel films were determined by phenolphthalein assay and acid–base titration. The films were characterized by solid state 13C NMR, ATR–FTIR, thermogravimetric analysis, and differential scanning calorimetric, and analyzed for tensile strength, swelling ratio, drug loading, release, hemocompatibility, in vitro cytotoxicity, and implantation test. An increase in the concentration of βCD in feed increased the active βCD content of the hydrogel films but reduced their extent of interpolymer crosslinking. The βCD‐g‐HEC hydrogel films with high active βCD content showed maximum drug loading whereas those with high crosslinking density were capable of controlling the drug release for long duration. Hemolysis assay and in vitro cytotoxicity study revealed the biocompatible nature of the hydrogel films whereas implantation test indicated their minimal inflammatory effect. From the overall results, βCD‐g‐HEC hydrogel films were found to be better alternative to the previously reported βCD‐HPMC and βCD‐CMC hydrogel films for enhanced loading and long‐term release, respectively, of the poorly soluble drugs. © 2018 Wiley Periodicals, Inc. The aim of present investigation was to characterize carboxymethyl tamarind gum (CMTG) based interpenetrating networks (IPNs) of aceclofenac for site specific sustained delivery. The drug loaded IPNs were prepared by using chitosan and CMTG as polymers and gluteraldehyde as crosslinking agent. The IPNs were characterized by Attenuated total reflectanceFourier transform infrared (ATR-FTIR) spectroscopy, thermal analysis, X-ray powder diffraction and solid state 13 C-nuclear magnetic resonance spectroscopy. The prepared IPNs were evaluated for the drug entrapment efficiency and equilibrium swelling. The drug release from IPNs was studied in 0.1NHCl for 2h followed by phosphate buffer pH 6.8 for further 10h and compared with commercial tablet. The results of ATR-FTIR and thermal analysis for blank IPNs indicated intercalation of polymeric chains of crosslinked CMTG and chitosan. The results of solid state characterization revealed that the aceclofenac is compatible with IPNs. Entrapment efficiency of IPNs was found to be increased with increase in crosslinker concentration as well as amount of CMTG. The equilibrium swelling study indicated pH dependent swelling of IPNs. The drug release by IPNs showed sustained release of aceclofenac upto 12h while commercial formulation showed fast release within 8h. From the results, it can be concluded that the IPNs of CMTG and chitosan has potential in development of site specific sustained drug delivery.
