Chromatographic Methods for Quantitative Determination of Ampicillin, Dicloxacillin and Their Impurity 6-Aminopenicillanic Acid

Abstract: Two accurate, precise and sensitive high-performance thin layer chromatography (HPTLC) and high-performance liquid chromatography (HPLC) methods were developed for assay of ampicillin (AMP) and dicloxacillin (DX) in the presence of their impurity, 6-aminopenicillanic acid (APA). Method (A) is HPTLC method; using silica gel HPTLC F254 plates as a stationary phase with methanol: chloroform: acetic acid (1:9: 0.2, by volume) as a developing system. All the bands were scanned at 220 nm. Method (B) is reversed phas… Show more

“…More intriguingly, AMOX and DIC were detected in their binary mixture using spectrophotometric methods [28][29][30][31][32] and chromatographic methods [33][34][35]. Ternary mixture of DIC, ampicillin, and 6-APA was analyzed by HPTLC [36]. Additionally, DIC, AMOX, and many antibacterial agents were analyzed in poultry feathers by UPLC method [37] However, the reported methods offered lower efficiency towards the separation of AMOX and DIC in the presence of 6-APA either in pure form or in the formulation.…”

“…The methods used in this experiment were developed through accessing other relevant stability-indicating studies available in the literature. [10][11][12] The final HPLC-UV method was validated for system suitability, linearity, accuracy, precision, and specificity. To evaluate system suitability, theoretical plates, tailing factor, and peak resolution were calculated using the ampicillin peak area and LCMS solutions software (v 3.0; Kyoto, Japan).…”

“…The evaluation is based on the SD of the response and the slope of the curves of the four studied pesticides, but using a visual non instrumental method regarding to ICH recommendations [15][16][17][18][19][20][21][22][23] for HPTLC method (Table 1).…”
