ChemInform Abstract: REACTIONS OF ACETALS OF L‐RHAMNOSE WITH ORGANOLITHIUM COMPOUNDS. NEW SYNTHESES OF MYCARAL, OLIVOMYCAL, AND METHY 2,3,6‐TRIDEOXY‐α‐L‐ERYTHRO‐HEX‐2‐ENOPYRANOSIDE

Abstract: Methyl-a-i mycarosid (6s) wird aus dem Didesoxy-ulosid 2 durch Grignard-Addition erhalten. Das Benzylidenrhamnosid l a reagiert mi! Methyllithium in einem Schritt zu Mycaral (4) und Olivomycal (5). Mi! Lithiumdiisopropylamid erhalt man hingegen aus 1 a den ungesattigten Zucker 3s. Reaclions of Acelals of i.-Rhnmnose with Lilhium-orgnnic Compounds New Syntheses of M y d . Olivomycd. and Methyl 2 , 3 ,~T r i d e o x y~~-~~o~Methyl 0-1 mycaroside (6a) is obtained from the dideoxy-uloside 2 by Grignard addition. T… Show more

“…Amino sugars and branched-chain sugars can also be readily accessed from intermediate 2 . Reductive amination yielded l -ristosaminide ( 25 )which features in the ristomycin members of the vancomycin antibiotic familyin 84% yield from 2 and in only three steps from 1 (previously six steps). ,, Similarly, methyl Grignard addition to 2 affords l -mycaroside in 55% yield and two steps overall (previously five steps). To the best of our knowledge, these methods provide the current best synthesis of each target, based on standard metrics of step count, yield, and atom economy. Anhydrosugar products are amenable to similar derivatizations; for example, aminosugars 27 and 28 were obtained via reductive amination from 9 and 11 , respectively (Figure D, bottom).…”
