ChemInform Abstract: ′CASCADE′‐ AND ′NONSKID‐CHAIN‐LIKE′ SYNTHESES OF MOLECULAR CAVITY TOPOLOGIES

Abstract: Ausgehend von Monoaminen (I) und Diaminen (V) werden in einer Kaskaden‐Reaktion durch Cyanethylierung und Reduktion zu Aminen polyfunktionelle Derivate (IV) bzw. (VI) erhalten.

“…First synthesized by Vogtle and co-workers in 1978, dendrimers are branched, spherical macromolecules widely used to encapsulate objects ranging from NPs to drugs. Poly(amidoamine) (PAMAM) dendrimers are a dendrimer subclass that consists of an alkyl-diamine core, amidoamine branches, and a vast range of peripheral functionalities (Scheme ). The size of PAMAM dendrimers depends on the number of reaction cycles carried out during the synthesis .…”

“…Dendritic molecules can be visualised as repetitive layers of multifunctional blocks of a protected and unprotected scheme of complimentary monomers typically resulting in a fractallike tree in which each incorporated layer serves as a platform for the successive layer (Tomalia and Cheng 2012). The first completely characterised repetitively branched and polyfuctional molecule was generated by Vogtle et al in 1978 from a protocol based on cycles of nucleophile amine addition to electron-poor cyanoalkene, followed by reduction of the cyano groups which in turn yields new amine moieties for further reactions (Buhleier et al 1978). Several other contributions like Vogtle's, particularly those form the research groups of Newkome, Denkwalter and Tomalia (Denkewalter et al 1982, Newkome et al 1985, Tomalia et al 1985 build upon experiments setting out some fundamental principles of molecular organisation pioneered by Flory in the middle of the 20th century (Flory 1942).…”
