The optimization of extraction by the Box–Behnken design of response surface methodology, chemical composition, antioxidant activity (AA), and total phenolic content (TPC) of fresh and cured betel leaf essential oil (BLEO) was reported. A quadratic polynomial equation developed by multiple regression analysis revealed that the models were highly significant with R2 = .98 and p < .0001. At optimal extraction conditions of extraction time (140 min), extraction temperature (100°C) and leaf to water ratio (1:1.5 g/L), 0.35% and 0.48% of fresh and cured BLEO yield were obtained. Thirty‐three and thirty volatile compounds were identified from fresh and cured BLEO by gas chromatography‐mass spectroscopy, accounting for 96.4% and 93.96% of the total oils, respectively. Eugenol, estragole, linalool, α‐copene, and anethole with different percentages were accounted to be major components. Fresh BLEO exhibited strong antioxidant effects on DPPH and TPC than cured leaf EO. Both the EOs explored that AA had a positive correlation with their phenolic contents. Practical applicationsThe present study explores the effect of optimized exhaustive hydrodistillate condition on the analysis of chemical composition and AA of essential oil (EO). The findings of optimized extraction embodied in the present research can be scaled up for commercial and industrial purposes. Both fresh and cured BLEO may be used in the food industry as a natural food preservative and flavoring agent. It can be also used for various herbal, medicinal, and therapeutic products. The main identified compounds such as eugenol, estragole, and linalool are the most valuable for food and pharmaceutical industries. Eugenol was found to be the highest amount in the extracted EO that can be used as a local anesthetic in dentistry as an ingredient in dental cement for temporary fillings.

Medicinal herbs are one of the imperative sources of drugs all over the world. Star anise (Illicium verum), an evergreen, medium-sized tree with star-shaped fruit, is an important herb with wide distribution throughout southwestern parts of the Asian continent. Besides its use as spice in culinary, star anise is one of the vital ingredients of the Chinese medicinal herbs and is widely known for its antiviral effects. It is also the source of the precursor molecule, shikimic acid, which is used in the manufacture of oseltamivir (Tamiflu ® ), an antiviral medication for influenza A and influenza B. Besides, several other molecules with numerous biological benefits including the antiviral effects have been reported from the same plant. Except the antiviral potential, star anise possesses a number of other potentials such as antioxidant, antimicrobial, antifungal, anthelmintic, insecticidal, secretolytic, antinociceptive, antiinflammatory, gastroprotective, sedative properties, expectorant and spasmolytic, and estrogenic effects. In this study, the in vitro spermostatic action, hemolytic action in combination of parabens of two essential oils (EOs) from Eugenia brejoensis Mazin (Myrtaceae, EbEO) and Myroxylon peruiferum L. (Fabaceae, MpEO) is reported, in addition to the first chemical characterization of MpEO. The EOs were obtained by hydrodistillation and characterized by gas chromatography-mass spectrometry (GC-MS). Different concentrations of both EOs (25 to 400 µg/mL) were mixed with human ejaculate and sperm motility was assessed after 5, 15 and 30 min. A total of 44 compounds were identified in MpEO; the major components were spathulenol, α-pinene, caryophyllene oxide, limonene and β-elemene. Both EOs exhibited spermostatic action in a dose-dependent manner. The best results were obtained for EbEO at all treatment times, showing IC 50 values of 454.75 µg/mL (5 min), 417.68 µg/mL (15 min) and 403.93 µg/mL (30 min). M. peruiferum also showed significant immobilizing potential with IC 50 values of 620.39 µg/mL (5 min), 652.70 µg/mL (15 min) and 579.37 µg/mL (30 min). Both oils showed low cytotoxicity against human erythrocytes. This is the first report on human spermostatic action for EOs from E. brejoensis and M. peruiferum which has antimicrobial activity and low cytotoxicity. These results suggest the need for further research on toxicological aspects of both EOs and their components in order to develop new pharmaceutical preparations using them as leader compounds.
