Chemical and immunochemical identification of the second major rabbit immunoglobulin λ chain polypeptide bearing c7 epitopes

Abstract: The purification of lambda chain-containing IgG fraction from pooled sera of Basilea rabbits, which were bred and selected for the expression of a high level of lambda chains positive for c7 but negative for c21, was carried out. On the basis of specific binding to anti-c7 antiserum, the c7 lambda chain fraction in serum IgG was shown to account for up to 70% of the total immunoglobulin light chains, the remaining 30%, bearing the expected k2 bas isotype. Peptide mapping of the mixed light chains (lambda + k2 … Show more

“…In addition to the initial 3 Mb gap, the scaffold containing the IGL genes consists of more than 100 contigs with gaps between them. Rabbit lambda constant region genes, IGLC5 paired with IGLJ5 and IGLC6 paired with IGLJ6 are known to be functional (Hayzer et al 1990; Jaton et al 1990). …”

“…This apparent contradiction can be explained if we assume that the in vitro expressed Ch6 region, in the absence of the V domain, may have failed to fold correctly, thus preventing its binding to anti-c7 antibody, or that the relative affinity of anti-c7 antisera for epitopic sites on the surface of the C domain may be too low to ensure adequate binding. In the accompanying report [39], we demonstrate that, indeed, the Ch6 sequence is characteristic of the c7 phenotype; hence, for the sake of clarity, we shall consider the gene Ch6 to be the gene encoding the C region of c7 h chains.…”
