Bacillus coagulans T1.2, an indigenous bacterium from shrimp ponds in East Lampung, has been found to reduce total ammonia nitrogen (TAN). The purpose of this study was to know the effectivity of immobilization of bacterial by sodium alginate from marine brown algae to reduce wastewater pollution from shrimp culture. The brown algae, Sargassum sp. and Padina sp., were collected from Pesisir Barat and Ketapang beach, Lampung. Alginate was extracted from both Sargassum and Padina by alkali method. The bacterial immobilization beads were made by mixed Bacillus coagulans suspension with alginate (1:3 v/v) and formed beads by 1,5 ml syringe without needle. In the in vitro study, the immobilized bacteria were submerged in artificial wastewater (2 beads ml-1) and incubated for 20 days. The control group includes the same bacteria without immobilization. The TAN was measured and the viability of bacteria was evaluated after the incubation period. The results showed that using alginate from Sargassum sp. and Padina sp. as a matrix of immobilization indigenous bacterial Bacillus coagulans effectively and significantly reduce the content of Total Ammonia Nitrogen (TAN) in wastewater. The viability of bacteria immobilized with the alginate of Sargassum sp. and Padina sp. better than the bacterial treatment without immobilization.

Vibrio vulnificus, Vibrio parahaemolyticus and Vibrio harveyi have been found in aquatic environments and suspected as the primary trigger of WFD (White Feces Disease) outbreaks in aquaculture. This Vibrio spp. has an antibiotic resistance to Ampicillin, Co-Amoxiclav, Amoxicillin, Azithromycin, Actinobacteria and Ciprofloxacin HCL. Actinobacteria and alginate have been reported to increase the marine biota resistance against diseases through prebiotic and probiotic mechanisms. This study aims to discover and increase the secondary metabolite production of Actinobacteria-Alginate and its ability as anti-vibrio. Alginate extraction in the samples dated September 2020 originally from Teluk Awur Bays, Jepara, Central Java, Indonesia (33.73±1.84%) was considerably higher than in May 2021 (22.67±0.3%). Samples were taken from sediment and mangrove root. Actinobacteria strains are macroscopically and microscopically similar to the genus Streptomyces. The most well-known antibiotics were produced by Streptomyces spp. The anti-vibrio test was carried out by Kirby-Bauer disc diffusion. The results were observed by measuring the inhibition zone surrounding the paper disc using a digital calliper. Co-culture strain 90 together with alginate have an approved antibacterial activity against all Vibrio spp. in the concentration of 10.disc-1 mg and 5 mg.disc-1. Co-culture Actinobacteria with alginate has remarkably changed the green-yellow color to olive green/dark red-orange (strains 3, 62, 63, 72, and 90), indicating the transformation of the formation alginate with pigments into other compounds through the biosynthetic pathway. Therefore, alginate enables to support of Actinobacteria by induction the active secondary metabolite as an anti-vibrio to counteract the bacterial pathogen diseases.

A preliminary study to gauge the antimicrobial potency of Xylocarpus granatum mangrove fruit extract and sodium alginate extract against pathogenic microbe from the species Pseudomonas aeruginosa and Staphylococcus epidermidis. Lotions made of mangrove fruit extract (Sample 1) lotion and of a mixture of fruit extract and sodium alginate extract lotion (Sample 2) were topically tested on incision cut wound on mice abdominal area. The lotion was applied daily for five consecutive days. Observation on the test subjects were conducted to determine the rate of blood agglutination, tissue recovery rate, and fibroblast development rate using histology. The results showed that X. granatum extract lotion displayed significant antimicrobial activity against both pathogenic microbe species and did not show any conflict with the microbial activity found in sodium alginate lotion. Lotion adhesiveness test measured sample 1 at 0.26 seconds and sample two at 0.16 seconds. Both samples were shown to be oily in water. Subject observation showed blood coagulation on the first day, onset of tissue recovery on the second day and by the third day the wound had undergone complete tissue recovery. Observation on the fifth day showed that fibroblast tissue on the subject with sample 2 treatment was more solid than that with sample 1 treatment. It was concluded that the mix of X. granatum mangrove fruit and sodium alginate extracts showed most potency in wound treatment.Key words: X. granatum; incision wound; lotions; skin; histology; fibroblast.
