Single‐spored isolates of Pyrenophora associated with spot and net type net blotch of barley were compared using total DNA banding patterns, morphological and cultural characteristics, symptomatology and mating studies. Isolates of spot and net type net blotch were found to vary regarding conidium length and cultural growth rate. Mating studies among and between ascospore, spot and net type isolates proved unsuccessful under the conditions studied. Total DNA polymorphisms of the net spot and ascospore isolates digested with the restriction enzymes HpaII and HaeIII showed that the isolates have similar banding patterns. Random amplified polymorphic DNA (RAPD) showed that the banding patterns of the spot and net type isolates were again similar but were distinct from outgroups such as P. semeniperda and P. triticirepentis. The homology in DNA banding patterns of local isolates indicated that the difference in conidium length is insufficient to separate them as two species. It is concluded that spot and net type isolates occurring in South Africa belong to P. teres. Therefore spot type lesions are caused by P. teres f. sp. maculata and not by P. japonica as reported previously.

A Calonectria sp. forming a Cylindrocladium anamorph was found to be commonly associated with leaf spot and cutting rot of Eucalyptus in Brazil, Colombia, Ecuador and Argentina. Based on morphology, isolates of this species resembled Cal. hederae, Ca/. reteaudiiand Cy. leucothoes, taxa that are commonly referred to as the Cal. reteaudii-complex. Using DNA sequence data of the ITS1, 5.8S and ITS2 of the rRNA gene, as well as the/9-tubulin gene, this species was shown to represent a previously described, but rather poorly known species, Cal. spathulata, for which an emended description is provided. Furthermore, the recently described Cy, perseae was also shown to be synonymous with Cy. leucothoes, a species shown to be distinct from Cy. spathulatum. Based on vesicle and conidium morphology, as well as perithecial colour and ascospore morphology, it appears that Cal. hederae and Cal. reteaudii should also be retained as separate species. Additional collections and cultures would be required, however, to suitably characterise the latter two species in future studies.

Hormographiella-like strains, isolated from different natural substrates and producing sclerotia and occasionally basidiomata of Coprinus cinereus, were compared morphologically and using molecular techniques with clinical strains of Hormographiella aspergillata and H. verticillata. Analysis of restriction fragment length polymorphisms of ribosomal and mitochondrial-like DNA confirmed interspecific differences between H. aspergillata and H. verticillata, supporting the morphological data, and helped demonstrate that H. aspergillata is the anamorph of C. cinereus. The latter was confirmed also by crossing tests. The analysis of the mtDNA restriction profiles revealed intraspecific variability in C. cinereus, which allowed differentiation of clinical and environmental strains. Due to the implication of C. cinereus and Hormographiella in human opportunistic infections, the antifungal susceptibility test is included. Results show that all strains were susceptible to miconazole, itraconazole and ketoconazole but not to flucytosine and fluconazol. Susceptibility against amphotericin B was variable; while H. verticillata was susceptible, four out of seven C. cinereus strains tested were resistant.
