Characterization of an antibacterial peptide produced by Brevibacterium linens

Abstract: Aims: The aim of this research was to investigate the antimicrobial activity produced by Brevibacterium linens ATCC 9175. Methods and Results: A bacteriocin produced by the red smear cheese bacterium B. linens ATCC 9175 was identi®ed. The antimicrobial activity was ®rst produced at the exponential growth phase. A crude bacteriocin obtained from the culture supernatant¯uid was inhibitory to some indicator strains. It inhibited the growth of Listeria monocytogenes ATCC 7644, B. linens ATCC 9172 and Corynebacteri… Show more

“…Antimicrobial activity was determined essentially as described by Motta and Brandelli (2002). An aliquot of 20 µl of BLS P34 (1600 AU) was applied on surface of BHI agar plates previously inoculated with a swab submerged in suspensions of E. faecalis or S. aureus that corresponded to a 76 GÉSSICA A. COSTA et al…”

“…The antibacterial activity was initially tested against Listeria monocytogenes ATCC 7466 by agar disk diffusion assay (21). Aliquots of 24-h culture media were centrifuged at 10,000 g for 10 min, at 4°C.…”

“…Plates were incubated aerobically at 35°C for 24 h. The diameters of the inhibition zones around the disks were measured. The activity of BLS was defined as the reciprocal of the highest dilution yielding a zone of growth inhibition, and expressed as activity units (AU) per mL (21).…”

“…Antimicrobial activity was determined essentially as described elsewhere [32]. Briefly, aliquots of 20 µl of purified peptide P34 and nisin were applied on cellulose disks (6 mm) on BHI agar plates previously inoculated with indicator strain suspension (L. monocytogenes ATCC 7644 or L. seeligeri AC 82/4), which corresponded to 0.5 McFarland turbidity standard solution.…”
