Characterization of a bovine herpesvirus type 4 isolated from the spinal cord of a cow with astasia

Abstract: A bovine herpesvirus (BHV) strain designated B11-41, was recently isolated from the spinal cord of a cow with astasia. By BHV type specific polymerase chain reaction (PCR), a BHV-4 specific fragment was amplified from the DNA of strain B11-41. The nucleotide sequence of the PCR product revealed high homologies (98.4% and 99.8%) with those of two BHV-4 strains (Movar 33/63 and 86-068). The BamHI restriction endonuclease cleavage patterns of the B11-41 were more similar to those of BHV-4 than those of other type… Show more

“…Viruses: Strain B11-41 of BoHV-4, isolated from the spinal cord of a cow with astasia, was used in this study [21,30]. The Los Angeles strain of bovine herpesvirus type 1 (BoHV-1) [18], preserved in the National Institute of Animal Health, Tsukuba, Japan, was used as a control to determine the specificity of the enzyme-linked immunosorbent assay (ELISA).…”

“…However, since gammaherpesviruses usually persist latently in lymphocytes, and since BoHV-4 has been isolated from a wide variety of clinical conditions in cattle, it is not evident that persistent infection in the nervous system is a common biological property of all BoHV-4 strains. Recently strain B11-41 of BoHV-4, which belongs to the DN 599-like group, was isolated from the spinal cord of a cow with astasia in Japan [21,30]. Our aims in the present study were to investigate the pathogenicity and tissue tropism of the virus in cattle.…”

“…Movar-like viruses have mainly been isolated in Europe, and DN 599-like viruses have mainly isolated in North America. In Japan, strain B11-41 of BoHV-4, which belongs to the DN 599-like group, was first isolated from the spinal cord of a cow with astasia [24]. Recently, a BoHV-4 strain was isolated from a mammary lesion of a cow with clinical mastitis from a farm in Fukushima prefecture in Japan [17].…”

“…DNA was extracted from the cell pellet using a commercially available test kit (SepaGene, Sanko Junyaku Co., Ltd., Tokyo, Japan) following the manufacturer's instructions, dissolved in a small volume of TE buffer (10 mM Tris-HCl, pH 8.0 and 1 mM EDTA) and then the DNA concentration was measured. Strain B11-41 [24], Movar 33/63 and 86-068 [2,18] were used as controls for BoHV-4. BoHV-1 (strain 758) [19] and BoHV-2 (Minnesota strain) [25] were also used as controls for bovine herpesviruses.…”

“…Other animal DNA viruses used in this study are: a Los Angeles strain of bovine herpesvirus 1 (BoHV-1) [11], a WC-11 strain of alcelaphine herpesvirus 1 (AlHV-1) [12], ovine herpesvirus 2 (OvHV-2) [13], an HH-1 strain of equine herpesvirus 1 (EHV-1) [8], a YS-81 strain of pseudorabies virus (PRV) [2], and a Fukuroi strain of bovine adenovirus 7 (BAdV-7) [7]. Additionally, DNA samples extracted from a Chiba strain of parapoxvirus (PPV) [9] and a B11-41 strain of bovine herpesvirus 4 (BoHV-4) [15] were provided by Dr. H. Sentsui, National Institute of Animal Health, Japan.…”
