Abstract: ABSTRACT. Three different polymerase chain reaction (PCR) protocols were evaluated for their ability to detect bovine herpesvirus 2 (BoHV-2): single-step PCR with 3 reaction stages (denaturation, annealing and extension), 2 reaction stages (denaturation and annealing/ extension; shuttle PCR), and semi-nested PCR with 3 reaction stages. We describe 3 outbreaks of superficial dermatitis caused by bovine herpesvirus 2 (BoHV-2) in dairy breed calves. Clinically, all of the affected calves were 12-26 d of age, had alopecia and crusts on the face and ears, and were non-pruritic and afebrile. Affected animals recovered spontaneously without any treatment within 2-4 wk after onset of clinical signs based on 1 herd with follow up. Histologic examination of all skin crust or tissue samples identified neutrophilic inflammation, mild hyperkeratosis, multinucleate syncytial cells, and intranuclear inclusion bodies in the syncytial cells. Real-time PCR testing on affected surface crusts or tissue provided evidence of BoHV-2, and testing, where performed, was negative for parapoxvirus including bovine papular stomatitis virus and the ovine form of malignant catarrhal fever tested in EDTA blood samples. Bovine viral diarrhea virus also was negative by ELISA, as well as bovine herpesvirus 1 by immunohistochemistry. Direct electron microscopy of infected tissues in the first outbreak revealed herpesvirus-like particles.

Two common viral diseases in psittacine birds including budgerigar fledgling disease (BFD), generally called avian polyomavirus (APV) infection, and psittacine beak and feather disease (PBFD) have similar clinical manifestations characterized by feather disorders. A duplex shuttle PCR was developed for detection of APV and PBFD virus (PBFDV). Two pairs of oligonucleotide primers were designed to amplify a 298‐bp fragment of the t/T antigen region of APV genome and a 495‐bp fragment of the capsid protein region encoded by open reading frame (ORF) C1 of PBFDV genome, respectively. In the present study, APV and PBFDV were detected simultaneously in one tube by duplex shuttle PCR using these two pairs of primers. The detection limits were 2 viral copies of APV and 3 viral copies of PBFDV. In the clinical application, we detected 16 APV‐positive, 15 PBFDV‐positive, and 3 mixed infected samples in 39 samples examined. Sequences of the amplified products were read. The t/T antigen region was conserved in the APV‐positive samples as expected. ORF C1 of PBFDV genome showed diversity. Phylogenic analysis indicated that PBFDV ORF C1 consisted of 6 clusters which were related to subfamilies of psittacine birds. Our duplex shuttle PCR could be a useful method for differential diagnosis and molecular epidemiology of BFD and PBFD.

Characterization of Bovine Herpesvirus Type 4 Isolated from Cattle with Mastitis and Subclinical Infection by the Virus among Cattle

ABSTRACT. An outbreak of contagious mastitis occurred among cattle on a farm, and bovine herpesviruses were isolated from the affected mammary tissues, scabs and abscess discharge of the cattle. A bovine herpesvirus type 4 (BoHV-4)-specific fragment was amplified from the isolates by polymerase chain reaction (PCR). Restriction endonuclease analyses demonstrated that the isolates were related to Movar-like European type BoHV-4. To determine the ratio of BoHV-4 subclinical infection in the cattle, a genomic survey was performed by PCR for cattle that were moved to the animal hygiene service station in Ibaraki prefecture. The BoHV-4 genome was occasionally detected in peripheral blood leukocytes, lymph nodes and nervous tissues. The rate of BoHV-4 subclinical infection was relatively high in the cattle.
