Development of a Shuttle Polymerase Chain Reaction for the Detection of Bovine Herpesvirus 2.

Abstract: ABSTRACT. Three different polymerase chain reaction (PCR) protocols were evaluated for their ability to detect bovine herpesvirus 2 (BoHV-2): single-step PCR with 3 reaction stages (denaturation, annealing and extension), 2 reaction stages (denaturation and annealing/ extension; shuttle PCR), and semi-nested PCR with 3 reaction stages. All the PCR protocols showed the same sensitivity (detection limit of 0.4 TCID 50 ). A non-specific band sometimes appeared in mock cell DNA at annealing temperatures below 64°C… Show more

“…PCR was essential to confirm the infectious agent in these cases, given its high specificity and sensitivity in detecting and identifying viral nucleic acid. 1,6,7,11 In one case, positivity for BoHV-2 virus was identified in a 3.5-y-old animal presented at a slaughterhouse. 20 The lesions characterized by lumps and scattered circular areas of alopecia were identified on the head, neck, shoulders, and on the perineum, representing the chronic pattern of PLSD.…”

“…and feline herpesvirus-1 in cats with upper respiratory tract disease (16). Shuttle PCR is another rapid method for detecting the target because annealing and extension are done simultaneously (10). Therefore, a combination of duplex PCR and shuttle PCR can make differential diagnosis easy.…”
