Cellular cytotoxic response induced by highly purified multi-wall carbon nanotube in human lung cells

Abstract: Carbon nanotubes, a promising nanomaterial with unique characteristics, have applications in a variety of fields. The cytotoxic effects of carbon nanotubes are partially due to the induction of oxidative stress; however, the detailed mechanisms of nanotube cytotoxicity and their interaction with cells remain unclear. In the present study, we focus on the acute toxicity of vapor-grown carbon fiber, HTT2800, which is one of the most highly purified multi-wall carbon nanotube (MWCNT) by high-temperature thermal t… Show more

“…Carbon nanotubes have greatly advanced these fields. Similarly as other nanoparticles, carbon nanotubes can be readily taken up by cells and are noncytotoxic within a certain concentration limit if suitably functionalized [25] [26]. The main finding of this study was that HTT2800 has a significant effect on intracellular lipid accumulation in C2C12 cells.…”

“…These results were in accordance with a previous report, in which a similar effect was observed in the BEAS-2 cell line. 23 By contrast, in the case of THP-1 cells, MWCNTs adhered around the cells, but were not ingested. This response was similar to that previously observed in U937 cells.…”

“…22 The objective of the present study was to confirm whether the expression of DJ-1 in U937 cells is universal. We recently also reported that exposure of BEAS-2B cells (derived from human bronchial epithelium) to HTT2800 proved cytotoxic, 23 causing arrest of proliferation. One of the most significant differences between U937 and BEAS-2B cells is their surface adhesiveness.…”

“…Similarly, exposure of nano-sized carbon black by pulmonary instillation exacerbated the atherosclerotic lesions in the aorta of mice susceptible to the development of AS 29) . A number of possible mechanisms have been proposed to explain the adverse health effects of MWCNTs, including their abilities to induce oxidative stress 15,30) and inflammation 31) , or induce platelet aggregation and vascular thrombosis 32) .
